Optimizing the Extraction of Keratin from Cattle Hoof Using Central Composite Design

Cattle hoofs are abundantly available by-product sources of organic material from the slaughterhouse. It can be successfully converted into keratin protein. Keratin extracted using alkali hydrolysis has a better conservancy structure. The purpose this study is optimizing extraction cuttle hoof. Designing experiment, analysis results, and optimization process parameters have been conducted by central composite design (CCD). Three factors (temperature (A), time (B), concentration NaOH (C)) each at five levels used to extract protein cattle hoof two response variables (dissolution % purity %) considered in study. results obtained demonstrate that temperature, concentration, showed significant effect on dissolution keratin. regression model shows all positive relation with percentage whereas only temperature negative percentage. observed Biuret Fourier-transform infrared spectroscopy (FTIR) tests preservation structure extractive FTIR spectra, indicates amide I II occur wave length 1633 1542 cm?1 for raw 1650 protein, respectively. Thus, it concluded could offer an alternative source. Finally, had optimum value 0.5 M NaOH, 60 minutes reaction time, 55°C 85% 89.6% purity.
