Osmotic Characteristics of Erythrocytes After Freezing with Composition-Modified Cryopreservatives

چکیده
Here, we have studied the osmotic parameters of erythrocytes frozen in composition-modified media with glycerol and 1,2-propanediol. A decreased concentration a cryopreservative an increased content impermeable protective component sorbitol were established to affect erythrocyte characteristics at different stages freezing hypothermic storage. Reduction 1,2-propanediol NaCl concentrations when augmenting sucrose agent enabled changing two-stage mode freeze-thawing single-stage one. The presented here modification cryoprotective medium allowed diminish level damage during freeze-thawing-washing out, that might prevent inflammation development body transfusing these cells.

Graphic analysis of osmotic fragility of erythrocytes.
A precise and highly reproducible method for analyzing the osmotic fragility of erythrocytes with a minute amount of blood (less than 10 mul) is described. The osmotic fragility curves are recorded with a coil planet centrifuge with accessories and a scanning photodensitometer. The recorded curves are transcribed by a DuPont Curve Resolver and their components are analyzed. Normal fragility cur...
متن کاملDimensional, Osmotic, and Chemical Changes of Erythrocytes
Previous studies have indicated that acid glucose-citrate solutions provide the best preservation available at the present time (1). The mixture proposed by Loutit and Mollison (2, 3), conventionally called ACD-I in the United States, has been most extensively studied and appears unsurpassed in its preservative qualities. It is added in amounts of 25 ml. per 100 ml. of blood. The study to be re...
متن کاملThe osmotic fragility of erythrocytes after prolonged liquid storage and after reinfusion.
Although it is recognized that red cells lose membrane during storage, estimation of the osmotic fragility of erythrocytes has not previously proven to be a useful measurement of the storage lesion. Erythrocytes from blood stored in CPD-A2 were found to have a markedly increased osmotic fragility. A major portion of this increase was found to be due to accumulation of lactate, which is only slo...
متن کاملOsmotic resistance of heat-damaged erythrocytes.
Whole blood was heated for twenty minutes at 40 degrees C., 45 degrees C., 50 degrees C., and 55 degrees C. Changes in the osmotic resistance of the heat-exposed cells were then determined. The values obtained were plotted as lysis increments. This treatment revealed groups of cells of varying heat sensitivity. After heating to 50 degrees C., cell groups were therefore prepared by partitioning ...
متن کاملFreezing characteristics and texture variation after freezing and thawing of four fruit types
One major problem with frozen fruits is a loss of texture. Therefore this study investigated the effects of the freezing process on the freezing profiles, texture, and drip loss of apple, mango, cantaloupe, and pineapple fruit samples. All frozenthawed fruits varied in these three properties because of diversity in the fresh fruits. Mango had the highest total soluble solids content and the low...
