Abstract Background Bispecific antibodies (bsAbs) and multispecific (msAbs) are a growing class of next generation biotherapeutics. However, multiple challenges, such as chain mispairing developability issues often associated with these complex modalities. Methods WuXi Biologics has established bsAb msAb engineering development platforms. By replacing CH1/CL domains one antibody Fab by corresponding T cell receptor (TCR) Cβ/Cα domains, WuXiBody® technology can ensure the correct heavy-light pairing. developed SDArBody™ platform, which utilizes single domain (VHH) building blocks to facilitate exploration more biologics, msAbs. Results shown in case studies, is able assemble regular mAbs ‘plug-and-play’ manner, that SDArBody® be utilized identify VHH leads then into Both platforms generate highly functional good developability. Conclusions The establishment could greatly meet needs biologics developers pursuit different biology therapeutic approaches through bsAbs
