Oxidative Stress Boosts the Uptake of Cerium Oxide Nanoparticles by Changing Brain Endothelium Microvilli Pattern

چکیده
Vascular oxidative stress is considered a worsening factor in the progression of Alzheimer’s disease (AD). Increased reactive oxygen species (ROS) levels promote accumulation amyloid-? peptide (A?), one main hallmarks AD. In turn, A? potent inducer stress. early stages AD, concomitant action and on brain capillary endothelial cells was observed to compromise blood–brain barrier functionality. this context, antioxidant compounds might provide therapeutic benefits. To aim, we investigated activity cerium oxide nanoparticles (CNP) human cerebral microvascular (hCMEC/D3) exposed oligomers. Treatment with CNP (13.9 ± 0.7 nm diameter) restored basal ROS hCMEC/D3 cells, both after acute or prolonged exposure A?. Moreover, found that extent uptake by +43% higher presence Scanning electron microscopy western blot analysis suggested changes microvilli structures cell surface, under pro-oxidant stimuli (A? H2O2), be involved enhancement uptake. This finding opens possibility exploit modulation pattern improve anti-oxidant particles designed counteract ROS-mediated cerebrovascular dysfunctions.

Background: The present study aimed to evaluate the effects of different concentrations of cerium oxide nanoparticles (CONPs) on the oxidative stress (OS) status in kidney, lung, and serum of rats. Methods: Male Wistar Rats were treated intraperitoneally with 15, 30, and 60 mg/kg/day of CONPs. The biochemical parameters, including total antioxidant capacity (TAC), total thiol group (TTG), malon...
متن کاملExperimental Study of Cerium Oxide Nanoparticles (CeNP) Against Malathion Induced Lung Oxidative Toxic Stress in Rats
Regardless of toxicity of nanoparticles, cerium oxide nanoparticles (CeNPs) are emerging as a multi-functional agent for biomedical purposes. On the other hand, Organophosphorus pesticides, like malathion, are inevitably found in the environment. The common involving pathway CeNPs and malathion share is oxidative stress. Therefore, we conducted this study to find the possible neutralizing or sy...
