چکیده
Abstract Study question Could the difference in controlled ovarian stimulation protocol between progestin-primed and GnRH-antagonist induce change expression genes of human granulosa cells? Summary answer The mitochondrial DNA (mtDNA) gene cells patients who underwent was significantly higher than GnRH-antagonist. What is known already (PPOS) has attracted attention many studies have reported similar pregnancy outcomes for both PPOS (GnRH-ant). However, our previous study demonstrated that rate lower compared to GnRH-ant (38th Hybrid Annual Meeting ESHRE, O-130). In this study, we evaluated (GCs) which affects reproductive outcome. Many shown a close association status GCs embryo’s quality. To date, with different protocols not been investigated. design, size, duration From November 2021 January 2022, all samples were obtained from provided written informed consent before inclusion cycles at single-institution. Single cell RNA-seq (scRNA-seq) performed on 2,224 corresponding MII-oocyte follicles total 16 normal reserve (aged < 40 years, AMH ≧1.1 ng/mL) undergoing chlormadinone acetate (n = 8), or cetrorelix 8). Participants/materials, setting, methods by follicular aspiration oocyte retrieval. fluid (FF) each follicle collected FF mixed together sequencing. isolated blood cellular debris using Percoll gradient centrifugation. There no significant distribution cluster. two GCs, identified 12 differentially expressed (DEGs). Interestingly, observed increase mtDNA as data confirmed qRT-PCR Limitations, reasons caution Our conclusions are limited due Asian need be validated across centers other ethnicities. Due small sample these should prompt further confirm findings. Wider implications findings These describe, first time, DEGs investigated COS single-cell level. This suggests progestin elevation mt may associated outcomes. Trial registration number Not applicable

P-60: The Effects of GnRH Antagonist (Cetrorelix) on Survival Rate of Ovarian Follicles in Anticancer Treatment Mice
Background: Infertility problem affects 20-30% of young couples. One of the known causes of spermatogenesis disorder is chemotherapy in patients with cancer which its side effect may last up to the end of the life. The aim of the present study is to investigate the effect of GnRH on viability of ovarian follicles defect produced by anticancer drug. Materials and Methods: In the present study ad...
متن کاملRegulation of Apoptosis and Steroidogenesis in Ovarian Preovulatory Granulosa Cells
Only a minute fraction of ovarian follicles present in a foetal ovary will complete the path to ovulation. The majority of the follicles will instead undergo atresia, a hormonally controlled apoptotic process. Apoptosis occurs throughout follicle development, with an extensive reduction in the number of follicles present at birth. Interestingly, this drastic reduction is not present among preov...
متن کاملAssessment of fertility protection and ovarian reserve with GnRH antagonist in rats undergoing chemotherapy with cyclophosphamide
BACKGROUND Reproductive function following chemotherapy is of increasing importance given that survival rates are improving. We assessed whether a gonadotropin-releasing hormone antagonist (GnRHant; cetrorelix) could promote ovarian protection against damage due to chemotherapy. METHODS Forty-two female Wistar rats were used in this study. Animals were divided into four groups: group I (n=9) ...

Objective(s): Pesticides has wide range of infertility in female reproductive. This study was done to evaluate the effect of movento pesticide on development of granulosa cells and ovarian follicles and FoxO1 and Vnn1 gene expression in BALB/c mice. Materials and Methods: In this study 40 healthy BALB/c mice 5-6 weeks age were used. Animals were randomly allocated into four groups. Control (wi...
متن کاملLetrozole+ GnRH antagonist stimulation protocol in poor ovarian responders undergoing intracytoplasmic sperm injection cycles: An RCT
BACKGROUND Gonadotropin-releasing hormone (GnRH) antagonist protocol has been proposed as a potentially proper option for the patients with limited ovarian reserve. Nevertheless, there is no significant difference in terms of clinical pregnancy between the GnRH antagonist and agonist cycles. The use of aromatase inhibitors such as letrozole was suggested by some studies. OBJECTIVE The object ...
