چکیده

Abstract Background The study of the intestinal epithelial barrier is hampered by lack a suitable in vitro model system. Patient-derived organoids (PDOs) recapitulate cellular diversity and patient heterogeneity provide relevant platform to epithelium its response microbial or inflammatory challenges. Here, we present characterization an bowel disease (IBD) organoid biobank derived from Crohn’s Ulcerative Colitis patients application for IBD drug development efforts. Methods PDOs different regions gastrointestinal tract 14 were established. To full diversity, specialized medium formulations defined that support differentiation distinct cell lineages as confirmed transcriptional profiles histology. inflammation driven loss integrity observed mucosa patients, employed monolayers mimicked environment cytokine challenge (TNF, IFNγ). Results In media, displayed gene expression patterns consistent with maturation types found human gut. robustly induces across seven patient-derived monolayers. breakdown was ameliorated all models treatment therapy Tofacitinib. Conclusion Intestinal generated capable differentiate into mature types. PDO-derived built trans-epithelial electrical resistance (TEER) are function mediated breakdown. Proof-of-concept studies Tofacitinib demonstrated suitability assay

Patient-derived xenografts: a relevant preclinical model for drug development

Identifying appropriate preclinical cancer models remains a major challenge in increasing the efficiency of drug development. A potential strategy to improve patient outcomes could be selecting the 'right' treatment in preclinical studies performed in patient-derived xenografts (PDXs) obtained by direct implants of surgically resected tumours in mice. These models maintain morphological similar...

development and implementation of an optimized control strategy for induction machine in an electric vehicle

in the area of automotive engineering there is a tendency to more electrification of power train. in this work control of an induction machine for the application of electric vehicle is investigated. through the changing operating point of the machine, adapting the rotor magnetization current seems to be useful to increase the machines efficiency. in the literature there are many approaches wh...

Establishment and characterization of an orthotopic patient-derived Group 3 medulloblastoma model for preclinical drug evaluation

Medulloblastomas comprise a heterogeneous group of tumours and can be subdivided into four molecular subgroups (WNT, SHH, Group 3 and Group 4) with distinct prognosis, biological behaviour and implications for targeted therapies. Few experimental models exist of the aggressive and poorly characterized Group 3 tumours. In order to establish a reproducible transplantable Group 3 medulloblastoma m...

Patient-Derived Xenograft (PDX) Models: An Emerging Platform for Cancer Drug Development and Translational Research

Previously, cancer drug development was an arena that relied almost exclusively on transgenic and severely compromised immuno-deficient (SCID) mice to measure tumor growth in response to novel therapeutic drugs. Development has now evolved to the point where several different murine models are now employed. The industry has expanded to include syngeneic and humanized mice, in addition to PDX mo...

Stem Cell-Derived Human Intestinal Organoids as an Infection Model for Rotaviruses

UNLABELLED Directed differentiation of stem cell lines into intestine-like tissue called induced human intestinal organoids (iHIOs) is now possible (J. R. Spence, C. N. Mayhew, S. A. Rankin, M. F. Kuhar, J. E. Vallance, K. Tolle, E. E. Hoskins, V. V. Kalinichenko, S. I. Wells, A. M. Zorn, N. F. Shroyer, and J. M. Wells, Nature 470:105-109, 2011). We tested iHIOs as a new model to cultivate and ...
