Perspectives in Hydrophobic Interaction Temperature- Responsive Liquid Chromatography (TRLC)

چکیده

Temperature-responsive liquid chromatography (TRLC) is an emerging green high performance (HPLC) mode allowing reversed phase-type separations while necessitating only water as the mobile phase. The columns therein are typically packed with silica particles to which stimuli-responsive polymers anchored. In hydrophobic interaction TRLC, such depict a loss of solubility when increasing temperature above characteristic conversion temperature, causing large changes in retention over quite narrow and mild ranges (~5–55 °C). TRLC circumvents concerns about analyte or column degradation that can occur implementing temperatures (>80 °C) on conventional reversed- phase columns. It allows for using often spiked additives used reversed-phase LC. Therefore, this separation greener, cheaper, isocratic analyses under non-denaturing conditions. absence compositional solvent gradients also exploitation combination refractive index detection. Purely aqueous mostly applicable solutes depicting 1 < LogP 5, yet these be expanded through implementation combined organic mobiles phases, preserving temperature-responsive effects. first installment, our recent developments, new possibilities, current limitations use 1-D discussed, described respect fundamentals HPLC.

Hydrophobic Interaction Chromatography.

Most proteins and large polypeptides have hydrophobic regions at their surface. These hydrophobic "patches" are due to the presence of the side chains of hydrophobic or nonpolar amino acids such as phenylalanine, tryptophan, alanine, and methionine. These surface hydrophobic regions are interspersed between more hydrophilic or polar regions and the number, size, and distribution of them is a sp...

High-temperature ultrafast liquid chromatography.

A novel liquid chromatographic system which enables high temperature ultrafast liquid chromatography (HTU-FLC) has been designed through the careful consideration of heat transfer, band broadening, and pressure drop. Studies of the effect of linear velocity on the HETP show that column efficiency at high velocity, especially of well-retained solutes, dramatically improves at higher temperatures...

Hydrophilic interaction liquid chromatography (HILIC) in proteomics

In proteomics, nanoflow multidimensional chromatography is now the gold standard for the separation of complex mixtures of peptides as generated by in-solution digestion of whole-cell lysates. Ideally, the different stationary phases used in multidimensional chromatography should provide orthogonal separation characteristics. For this reason, the combination of strong cation exchange chromatogr...
