Phase Separation and Protein Partitioning in Compartmentalized Cell-Free Expression Reactions

Liquid-liquid phase separation (LLPS) is important to control a wide range of reactions from gene expression protein degradation in cell-sized space. To bring better understanding the compatibility such phase-separated structures with synthesis, we study emergent LLPS cell-free transcription-translation (TXTL) reaction. When TXTL reaction composed many proteins concentrated, uniformly mixed state becomes unstable, and membrane-less phases form spontaneously. This droplet formation induced when enclosed water-in-oil emulsion droplets, which water evaporates surface. As droplets shrink, smaller appear inside coalesce into large domains that partition localization synthesized reporter proteins. The presence PEG not only for versatile synthesis but also two capable partitioning. Our results may shed light on dynamic interplay toward construction synthetic organelles.
