چکیده
Background: Autoantibodies targeting a muscle-specific autoantigen, four-and-a-half-LIM-domain 1 (FHL1), have been previously identified in patients with idiopathic inflammatory myopathies (IIM) (1). Objectives: The aim of this project was to determine the prevalence and associations anti-FHL antibody South Australian histologically-confirmed IIM an autoimmune disease control (systemic sclerosis (SSc)). Methods: Sera from (n=267) Myositis Database (SAMD), SSc (n=174) Scleroderma Cohort Study (ASCS) followed at Royal Adelaide Hospital, healthy controls (HC, n=100) were analyzed for anti-FHL1 autoantibodies by Enzyme-Linked ImmunoSorbent Assay (ELISA). Clinical, serological histological details retrieved SAMD ASCS. Results: FHL1 more frequent (55/267, 20.5%) compared (18/174, 10%) (p<0.001) HC (4/100, 4%) (p<0.001). Muscular vessel inflammation atrophy seen frequently + - (p<0.01 p<0.05). Dysphagia, marked muscle atrophy, high CK levels inclusion body myositis (IBM) immune-mediated necrotizing myopathy (IMNM). In 35/54 patients, there no other myositis-specific present. Anti-FHL1 associated gastric antral vascular ectasia. Conclusion: detected 20.5% patients. IBM IMNM, presence anti-FHL1-autoantibodies severe as suggested dysphagia atrophy. References: [1]Albrecht I, Wick C, Hallgren A, Tjarnlund Nagaraju K, Andrade F, et al. Development of autoantibodies against muscle-specific FHL1 in severe inflammatory myopathies.
Mutations of the gene encoding four-and-a-half LIM domain 1 (FHL1) are the causative factor of several X-linked hereditary myopathies that are collectively termed FHL1-related myopathies. These disorders are characterized by severe muscle dysfunction and damage. Here, we have shown that patients with idiopathic inflammatory myopathies (IIMs) develop autoimmunity to FHL1, which is a muscle-speci...
متن کاملAutoantibodies in Idiopathic Inflammatory Myopathies
Autoimmune response to nuclear and cytoplasmic autoantigens is detected in about 60-80% of patients affected with idiopathic inflammatory myositis such as polymyositis (PM), dermatomyositis (DM) and inclusion body myositis (IBM). Some of the serum autoantibodies are shared with other autoimmune diseases (myositis-associated antibodies MAA) and some of them are unique to myositis (myositis-speci...

[Myositis specific and myositis associated autoantibodies in idiopathic inflammatory myopathies: a serologic study of 46 patients].
OBJECTIVE To characterize serum autoantibody profiles of patients with idiopathic inflammatory myopathies (IIM) by searching for myositis-specific (MSA) and myositis-associated (MAA) antibodies with sensitive and specific laboratory tests. METHODS We tested the sera from 46 Caucasian patients diagnosed as affected with IIM at the Division of Rheumatology of Padova University (21 polimyositis,...
متن کاملAbsence of Four-and-a-Half LIM Domain Protein 2 Decreases Atherosclerosis in ApoE-/- Mice.
OBJECTIVE Four-and-a-half LIM domain protein-2 (FHL2) is expressed in endothelial cells, vascular smooth muscle cells, and leukocytes. It regulates cell survival, migration, and inflammatory response, but its role in atherogenesis is unknown. APPROACH AND RESULTS To investigate the role of FHL2 in atherosclerosis, FHL2-deficient mice were crossed with ApoE-deficient mice, to generate ApoE/FHL...
