Pre-mRNA splicing in the new millennium.

The past year has witnessed refinements in models of spliceosome assembly pathways and in the understanding of how splicing factors of the serine/arginine-rich (SR) protein family function. The role of splicing in human genetic diseases has also received a lot of attention recently as exonic splicing enhancers become better understood.

RNAcatalyses nuclear pre-mRNA splicing

In nuclear pre-messenger RNA splicing, introns are excised by the spliceosome, a dynamic machine composed of both proteins and small nuclear RNAs (snRNAs). Over thirty years ago, after the discovery of self-splicing group II intron RNAs, the snRNAswere proposed to catalyse splicing. However, no definitive evidence for a role of either RNAor protein in catalysis by the spliceosome has been repor...

Organization of RNA polymerase II transcription and pre-mRNA splicing within the mammalian cell nucleus.

Introduction Splicing is an essential step in the processing of intron-containing pre-mRNAs before their transport out of the nucleus (for a review see [ 13). During the last few years, a significant number of studies have addressed the relationship of pre-mRNA splicing to nuclear structure (for a review see [Z]). Small nuclear ribonucleoprotein particles (snRNPs) as well as several non-snRNP s...

Pre-mRNA splicing imprints mRNA in the nucleus with a novel RNA-binding protein that persists in the cytoplasm.

We describe a novel RNA binding protein, Y14, a predominantly nuclear nucleocytoplasmic shuttling protein. Interestingly, Y14 associates preferentially with mRNAs produced by splicing but not with pre-mRNAs, introns, or mRNAs produced from intronless cDNAs. Y14 associates with both nuclear mRNAs and newly exported cytoplasmic mRNAs. Splicing of a single intron is sufficient for Y14 association....

Pre-mRNA splicing and retinitis pigmentosa.

Retinitis pigmentosa (RP) is a group of genetically and clinically heterogeneous retinal diseases and a common cause of blindness. Among the 12 autosomal dominant RP (adRP) genes identified, four encode ubiquitously expressed proteins involved in pre-mRNA splicing, demonstrating the important role that pre-mRNA splicing plays in the pathogenesis of retinal degeneration. This review focuses on r...
