PSXIV-15 Synergistic Inhibition of Staphylococcal Growth by Select Nitrocompounds and Sodium Chlorate

چکیده
Abstract Staphylococci are important mastitic-pathogens of humans and livestock. Chlorate is reported to be a potent inhibitor against staphylococci but adaptation by the microbes can occur. Presently, we cultured multi-drug resistant Staphylococcus aureus strain ATCC 12600 anaerobically at 39oC in nitrate-supplemented (5 mM) Tryptic Soy broth. The broth was treated without (control) or alone with 5 mM sodium chlorate (ClO3) 9 either 2-nitro-1-propanol (NPOH), ethyl nitroacetate (ENA) 2-nitropropionate (E2NPA) combinations CLO3 respective nitrocompound. Control cultures were incubated triplicate growth measured via absorbance (600 nm) during h. An analysis variance revealed an inhibitory effect treatments (P < 0.05; SEM = 0.078) on mean specific rates S. aureus, logarithmic being most rapid controls ENA EN2PA (1.45, 1.23 1.21 h-1, respectively) more than 92% slower NPOH NPOH+CLO3-treated (0.09 0.03 respectively). Growth solely (0.66 h-1) 50% decreased further plus E2NPA (0.39 0.45 h-1). After h growth, maximum optical densities (maxOD) highest for (0.52, 0.52 0.50 maxOD, respectively), intermediate (0.21 maxOD) lower yet ENA, (0.15, 0.11 0.02 maxOD did not differ from combination CLO3+NPOH. Results demonstrate marked synergistic prolonging effects combing no additional anti-staphylococcal activity NPOH.

Inhibitory effect of select nitrocompounds on growth and survivability of Listeria monocytogenes in vitro.
We report the effects of 2-nitro-1-propanol (2NPOH), 2-nitroethanol (2NEOH), and nitroethane (NE) on growth and survivability of Listeria monocytogenes. In all cases, inhibition was greatest with 2NPOH and least with NE. For example, specific growth rates of L. monocytogenes strain 18 declined (P < 0.05) 76, 60, and 29% from controls during aerobic culture at 37 degrees C in brain heart infusio...

Antibiotic therapies to eradicate medical device-associated infections often fail because of the ability of sessile bacteria, encased in their exopolysaccharide matrix, to be more drug resistant than planktonic organisms. In the last two decades, several strategies to prevent microbial adhesion and biofilm formation on the surfaces of medical devices, based mainly on the use of antiadhesive, an...

Background and Aims: Candidiasis is a fungal infection caused by Candida albicans. Uncontrolled use of antibiotics has resulted in drug resistance. This study has been conducted to find a suitable alternative for these drugs. Materials and Methods: Primarily, standard and clinical isolates of C.albicans were collected. In order to indentify clinical isolates from lambs, the conventional mycolo...

Interleukin-15 is a pleiotropic cytokine that has potential for cancer immunegene therapy. We previously reported the construction and characterization of IL-15 overexpression vectors pHi2-IL15-CMV-tat (L1) and pHi2-spIL15-CMV-tat (L3), as well as carcinoembryonic antigen promoter-amplified IL-15 expression plasmid vectors pHi2-IL15-CEA-tat (L2) and pHi2-spIL15-CEA-tat (L4). In the current stud...
