چکیده

Objective: Objective of the study was to standardize and evaluate suitability Single Base Extension (SBE) singleplex assay for rapid routine monitoring genetic homogeneity virus samples, from productivity improvement experiments in Mumps vaccine. A is a simple, cost effective tool quick identification single nucleotide polymorphism (SNP) at three known sites vaccine genome. The can be developed as or multiplex version with potential detection up ten SNP bases reaction. Methods: SNaPshot kit Applied Biosystems (ThermoFisher) used identify Approximate sizing locations individual primers were determined using Primer Focus Kit, also Biosystems. Genetic analyzer 3500 capillary electrophoresis processed samples. Results obtained data collection software analyzed GeneMapper software. Findings: Nucleotide G identified 1073 11345 C location 5261 all tested mumps thus demonstrating consistency hotspots experimental Conclusion: SBE found reliable relatively inexpensive multiplexing that would reduce time even further.

A microsphere-based assay for multiplexed single nucleotide polymorphism analysis using single base chain extension.

A rapid, high throughput readout for single-nucleotide polymorphism (SNP) analysis was developed employing single base chain extension and cytometric analysis of an array of fluorescent microspheres. An array of fluorescent microspheres was coupled with uniquely identifying sequences, termed complementary ZipCodes (cZipCodes), which allowed for multiplexing possibilities. For a given assay, que...

Single Nucleotide Polymorphism Analysis of Protamine Genes in Infertile Men

Background Single nucleotide polymorphism (SNPs) are considered as one of the underlying causes of male infertility. Proper sperm chromatin packaging which involves replacement of histones with protamines has profound effect on male fertility. Over 20 SNPs have been reported for the protamine 1 and 2. MaterialsAndMethods The aim of this study was to evaluate the frequency of two previously repo...

A single nucleotide polymorphism assay for the identification of unisexual Ambystoma salamanders.

Unisexual (all female) salamanders in the genus Ambystoma are animals of variable ploidy (2N-5N) that reproduce via a unique system of 'leaky' gynogenesis. As a result, these salamanders have a diverse array of nuclear genome combinations from up to five sexual species: the blue-spotted (A. laterale), Jefferson (A. jeffersonianum), smallmouth (A. texanum), tiger (A. tigrinum) and streamside (A....

A Microfluidic Device for Detection of Single Nucleotide Polymorphisms by Allele Specific Single Base Extension

We present a microfluidic device with integrated temperature sensors and heaters for single nucleotide polymorphism (SNP) detection. The allele specific primer is immediately annealed next to the polymorphic site, and extended by a single base. The extension product is then purified by solid phase capture, and subsequently released via chemical cleavage. After desalting, the released product is...
