Repair of DNA double-strand breaks in RNAPI- and RNAPII-transcribed loci

DNA double-strand breaks (DSBs) are toxic lesions triggered not only by environmental sources, but also a large number of physiological processes. Of importance, endogenous DSBs frequently occur in genomic loci that transcriptionally active. Recent work suggests occurring transcribed handled specific pathway(s) entail local transcriptional repression, chromatin signaling, the involvement RNA species and DSB mobility. In this Graphical Review we provide an updated view “Transcription-Coupled Repair” (TC-DSBR) mounted at Polymerase I (RNAPI) or II (RNAPII), highlighting differences common features, as well yet unanswered questions.
