Protein translocation pathways across the inner and outer mitochondrial membranes

Mitochondria import different proteins that are encoded by nuclear genes and synthesized in the cytosol. Separate translocases in inner and outer mitochondrial membranes like TOM, TIM23, TOB/SAM, TIM22 complex facilitate recognition, import and intramitochondrial sorting of preproteins. Various cytosolic factors as Hsp70 and auxillary factors assist in targeting these preproteins to their desti...

Tim50 Is a Subunit of the TIM23 Complex that Links Protein Translocation across the Outer and Inner Mitochondrial Membranes

Based on the results of site-specific photocrosslinking of translocation intermediates, we have identified Tim50, a component of the yeast TIM23 import machinery, which mediates translocation of presequence-containing proteins across the mitochondrial inner membrane. Tim50 is anchored to the inner mitochondrial membrane, exposing the C-terminal domain to the intermembrane space. Tim50 interacts...

Protein translocation across membranes.

Cellular membranes act as semipermeable barriers to ions and macromolecules. Specialized mechanisms of transport of proteins across membranes have been developed during evolution. There are common mechanistic themes among protein translocation systems in bacteria and in eukaryotic cells. Here we review current understanding of mechanisms of protein transport across the bacterial plasma membrane...

Structural Basis of Type 2 Secretion System Engagement between the Inner and Outer Bacterial Membranes

Sophisticated nanomachines are used by bacteria for protein secretion. In Gram-negative bacteria, the type 2 secretion system (T2SS) is composed of a pseudopilus assembly platform in the inner membrane and a secretin complex in the outer membrane. The engagement of these two megadalton-sized complexes is required in order to secrete toxins, effectors, and hydrolytic enzymes. Pseudomonas aerugin...

On the translocation of proteins across membranes.

Many proteins of intracellular organelles are first synthesized in the cytoplasm and are then specifically transferred across the membranes of the organelles. On the assumption that these transfers all occur by the same basic mechanism, we enumerate the rather stringent requirements that the mechanism must satisfy. A unitary molecular mechanism is then proposed that meets these requirements.
