چکیده

RUBylation plays essential roles in plant growth and development through regulating Cullin-RING ubiquitin E3 ligase (CRL) activities the CRL-mediated protein degradations. However, function of kernel remains unclear. Through genetic molecular analyses a small 501 (smk501) mutant maize (Zea mays), we cloned smk501 gene, revealed its function, defined pathway seed development. Smk501 encodes activating enzyme E1 subunit ZmECR1 (E1 C-TERMINAL RELATED 1) protein. Destruction by mutation resulted less embryo endosperm cell number smaller size. The transcriptome proteome profiling, hormone evaluation proliferation observation that disturbing expression mainly affects pathways on signal transduction, cycle progression starch accumulation during In addition, zmaxr1 (Auxin resistant 1), another RUB subunit, also showed similar defects Double zmecr1 lead to empty pericarp phenotype. is novel regulatory affecting development, majorly functions modifying multiple cellular progresses.

E1 Ubiquitin-Activating Enzyme UBA-1 Plays Multiple Roles throughout C. elegans Development

Poly-ubiquitination of target proteins typically marks them for destruction via the proteasome and provides an essential mechanism for the dynamic control of protein levels. The E1 ubiquitin-activating enzyme lies at the apex of the ubiquitination cascade, and its activity is necessary for all subsequent steps in the reaction. We have isolated a temperature-sensitive mutation in the Caenorhabdi...

Mathematical model for the ubiquitin activating enzyme E1

The ubiquitin-activating enzyme E1 (EC 6.3.2.19) represents the first step in the degradation of proteins by the ubiquitin proteasome pathway. E1 transfers ubiquitin from the ubiquitinated E1 to the ubiquitin carrier proteins (E2), ubiquitin-protein ligases (E3) and proteins. This process is rather complex, and known from the work of Haas, Ciechanover, Hershko, Rose and others. The occurrence o...

Evolution of the Ubiquitin-Activating Enzyme Uba1 (E1)

Ubiquitin tags diseased proteins and initiates an enzyme conjugation cascade, which has three stages. The first-stage enzyme Uba1 (E1) has evolved only modestly from slime mold to humans, and is > 14 times larger than Ub. Here we use critical point thermodynamic scaling theory to connect Uba1 (E1) evolution from yeast and slime mold to fruit flies and humans to subtle changes in its amino acid ...

Largazole and Its Derivatives Selectively Inhibit Ubiquitin Activating Enzyme (E1)

Protein ubiquitination plays an important role in the regulation of almost every aspect of eukaryotic cellular function; therefore, its destabilization is often observed in most human diseases and cancers. Consequently, developing inhibitors of the ubiquitination system for the treatment of cancer has been a recent area of interest. Currently, only a few classes of compounds have been discovere...

Homology Modelling of Human E1 Ubiquitin Activating Enzyme.

Human E1 is a key player in protein ubiquitination, however the E1 structure is not available. In this paper, we describe the derivation of a human E1 structure using molecular modelling based on the crystal structure of S. cerevisiae E1 and M. Musculus E1. Key interactions between our E1 model and ubiquitin are also discussed.
