Strain-level Identification of Beneficial Lactobacilli of Dairy Origin using 16S rRNA Sequencing: A Biotechnology Approach

Aim: This study investigated the cultural method and 16S rRNA gene analysis to reveal composition diversity of lactic acid bacteria (LAB) from dairy origin (spontaneous fermented cow milk) in Nigeria.
 Methods: Six samples which includes two raw milk, goat milk (nono) were collected subjected standard microbiological investigation using both molecular methods. The cultured on MRS media, isolates identified physiological biochemical parameters. DNA four selected probiotic LAB nono amplified PCR while amplicons electrophoresed agarose gel, pre-stained with ethidium bromide characterized by analysis. result sequencing analyzed NCBI BLAST. 
 Results: A total 55 presumptive isolated. Twenty nine (29) Lactobacillus plantarum representing 52.7%, Pediococcus acidilactici 15(27.2%), casei brevis 4(7.3%) fermentum 3(5.5%) recorded respectively. Result gel electrophoresis revealed size approximately 1500bp. used this confirmed as N17, N24, N10 N1 based sequence phylogenetic a 98-100% similarity high homology level affirms strain organism.
 Conclusion: has demonstrated existing known could be harnessed valuable sources for isolation potential organisms.
