چکیده
Lung surfactant (LS) is an outstanding example of how a highly regulated and dynamic membrane-based system has evolved to sustain wealth structural reorganizations in order accomplish its biophysical function, as it coats stabilizes the respiratory air-liquid interface mammalian lung. The present review dissects complexity structure-function relationships LS through updated description lipid-protein interactions membrane structures that synthesis, secretion, interfacial performance recycling. We also revise current models techniques employed study membranous architecture LS. It important consider structure functional properties are often studied bulk or under static conditions, spite function strongly connected with behaviour, sustained by very polymorphic lipid-lipid, protein-protein reorganize precise spatio-temporal coordinates. have tried underline evidences available existence such dynamism A last aspect synthesis assembly intracellular process ensure establishment proper driving surface activity, while protecting integrity other cell membranes. use simplified lipid partial natural materials purified from animal tissues could be too simplistic understand true molecular mechanisms defining vivo. In this line, we will bring into attention reader methodological challenges questions still open at full biological relevance.

Bacterial osmosensing: roles of membrane structure and electrostatics in lipid-protein and protein-protein interactions.
Bacteria act to maintain their hydration when the osmotic pressure of their environment changes. When the external osmolality decreases (osmotic downshift), mechanosensitive channels are activated to release low molecular weight osmolytes (and hence water) from the cytoplasm. Upon osmotic upshift, osmoregulatory transporters are activated to import osmolytes (and hence water). Osmoregulatory ch...
متن کاملLipid-protein interactions alter line tensions and domain size distributions in lung surfactant monolayers.
The size distribution of domains in phase-separated lung surfactant monolayers influences monolayer viscoelasticity and compressibility which, in turn, influence monolayer collapse and set the compression at which the minimum surface tension is reached. The surfactant-specific protein SP-B decreases the mean domain size and polydispersity as shown by fluorescence microscopy. From the images, th...
متن کاملRole of surfactant protein A (SP-A)/lipid interactions for SP-A functions in the lung.
Surfactant protein A (SP-A), an oligomeric glycoprotein, is a member of a group of proteins named collectins that contain collagen-like and Ca(2+)-dependent carbohydrate recognition domains. SP-A interacts with a broad range of amphipathic lipids (glycerophospholipids, sphingophospholipids, glycosphingolipids, lipid A, and lipoglycans) that are present in surfactant or microbial membranes. This...

Complex I11 (ubiquinol-cytochrome c reductase) was purified from beef heart mitochondria in the form of protein-phospholipid-Triton X100 mixed micelles (about 1:SO:lOO molar ratio). Detergent may be totally removed by sucrose density gradient centrifugation, and the resulting lipoprotein complexes retain full enzyme activity. In order to understand the role of surfactant in the mixed micelles, ...
متن کاملStructural characterization of human and bovine lung surfactant protein D.
Human and bovine surfactant proteins D (SP-D) were purified from late amniotic fluid and bronchioalveolar lavage on the basis of its Ca(2+)-dependent affinity for maltose. The molecular mass of a trimeric subunit was determined by matrix-assisted laser desorption ionization MS to lie in the range 115-125 kDa for human SP-D and 110-123 kDa for bovine SP-D. A single polypeptide chain was determin...
