چکیده
Aspergillus westerdijkiae, known as the major ochratoxin A (OTA) producer, usually occurs on agricultural crops, fruits, and dry-cured meats. Microorganisms produce OTA to adapt high osmotic pressure environment that is generated during food processing storage. To investigate relationship between biosynthesis osmolarity glycerol (HOG) pathway, transcription factor AwHog1 gene in A. westerdijkiae was functionally characterised by means of a loss-of-function mutant. Our findings demonstrated growth production mutant lacking decreased significantly more sensitive media. The ΔAwHog1 displayed lower rate 73.16% reduction wheat medium compared wild type. After three days culture, with 60 g/L NaCl 150 glucose slowed down 19.57% 13.21%, respectively. Additionally, expression genes reduced deletion gene. infection ability decreased, scab diameter pear 6% smaller than These data revealed plays key role response, growth, production, pathogenicity westerdijkiae.

The Stress Response Regulator AflSkn7 Influences Morphological Development, Stress Response, and Pathogenicity in the Fungus Aspergillus flavus
This study focused on AflSkn7, which is a stress response regulator in the aflatoxin-producing Aspergillus flavus. The ΔAflSkn7 mutants exhibited partially defective conidial formation and a complete inability to generate sclerotia, indicating AflSkn7 affects A. flavus asexual and sexual development. The mutants tolerated osmotic stress but were partially susceptible to the effects of cell wall...
متن کاملMonitoring Response of a Few bZip Transcription Factors in Response to Osmotic Stress in Sunflower
Background: Sunflower (Helianthus annuus L.) is one of the important vegetable oil supplies in the world and in Iran, as well. It is classified as a drought semi-tolerant crop; however, its yield is adversely affected by drought stress. Understanding the initial events in sensing stress and the related physiologic and biochemical events thereafter, is crucial in designing droug...
متن کاملAspergillus steynii and Aspergillus westerdijkiae as potential risk of OTA contamination in food products in warm climates.
Aspergillus steynii and Aspergillus westerdijkiae are the main ochratoxin A (OTA) producing species of Aspergillus section Circumdati. Due to its recent description, few data are available about the influence of ecophysiological factors on their growth and OTA production profiles. In this work, the effect of temperature (20, 24 and 28 °C) and water activity (aw) (0.928, 0.964 and 0.995) on grow...
متن کاملThe Master Transcription Factor mtfA Governs Aflatoxin Production, Morphological Development and Pathogenicity in the Fungus Aspergillus flavus.
Aspergillus flavus produces a variety of toxic secondary metabolites; among them, the aflatoxins (AFs) are the most well known. These compounds are highly mutagenic and carcinogenic, particularly AFB₁. A. flavus is capable of colonizing a number of economically-important crops, such as corn, cotton, peanut and tree nuts, and contaminating them with AFs. Molecular genetic studies in A. flavus co...
متن کاملOsmotic stress-coupled maintenance of polar growth in Aspergillus nidulans.
Free-living cells monitor extracellular 'osmotic strength' and respond metabolically to offset unfavourable osmotic intracellular solute concentrations. Here, we report the reconstruction of the Aspergillus nidulans salt stress-controlling MAP kinase pathway, based on homology analysis with known yeast genes. In A. nidulans, salt stress HOG genes, such as pbsA, hogA, ptpA and msnA, are upregula...
