Triple-Knockout, Synuclein-Free Mice Display Compromised Lipid Pattern

چکیده
Recent studies have implicated synucleins in several reactions during the biosynthesis of lipids and fatty acids addition to their recognised role membrane lipid binding synaptic functions. These are among aspects decreased synuclein functions that still poorly acknowledged especially regard pathogenesis Parkinson’s disease. Here, we aimed add existing knowledge deficiency (i.e., lack all three family members), with respect changes plasma, liver, two brain regions triple synuclein-knockout (TKO) mice. We describe long-chain polyunsaturated (LCPUFA) palmitic acid liver reduced triacylglycerol (TAG) accumulation non-esterified plasma free In midbrain, observed counterbalanced relative concentrations phosphatidylcholine (PC) cerebrosides (CER). also recorded a notable reduction ethanolamine plasmalogens midbrain mice, which is an important finding since abnormal ether metabolism usually associated neurological disorders. summary, our data demonstrates results alterations PUFA synthesis, storage CER, those polar principal compounds rafts many tissues. An ablation members causes more profound than previously shown be γ-synuclein alone. Possible mechanisms by may govern reported modifications TKO mice proposed discussed.

Inflammatory stress exacerbates lipid-mediated renal injury in ApoE/CD36/SRA triple knockout mice.
Both lipids and inflammation play important roles in the progression of kidney disease. This study was designed to investigate whether inflammation exacerbates lipid accumulation via LDL receptors (LDLr), thereby causing renal injury in C57BL/6J mice, apolipoprotein E (ApoE) knockout (KO) mice, and ApoE/CD36/scavenger receptor A triple KO mice. The mice were given a subcutaneous casein injectio...
متن کاملα-Synuclein leaves autophagy feeling compromised
I f the spindle isn't aligned properly, the spindle position checkpoint (SPOC) pauses mitosis so that the cell can make adjustments. Caydasi et al. identify a master switch for the checkpoint. When a yeast cell begins to bud, the SPOC detects whether the spindle has lined up parallel to the bud–mother cell axis. One of the main SPOC proteins is the kinase Kin4. What controls its activity is unc...
متن کاملAbnormal hematopoietic phenotypes in Pim kinase triple knockout mice
BACKGROUND Pim (proviral insertion in murine lymphoma) kinases are a small family of constitutively active, highly conservative serine/threonine oncogenic kinases and have 3 members: Pim1, Pim2, and Pim3. Pim kinases are also implicated in the regulation of B- and T- cell responses to cytokines and hematopoietic growth factors. The roles of Pim kinases in the regulation of primitive hematopoiet...
متن کاملFemale preproenkephalin-knockout mice display altered emotional responses.
The endogenous opioid system has been implicated in sexual behavior, palatable intake, fear, and anxiety. The present study examined whether ovariectomized female transgenic preproenkephalin-knockout (PPEKO) mice and their wild-type and heterozygous controls displayed alterations in fear and anxiety paradigms, sucrose intake, and lordotic behavior. To examine stability of responding, three squa...
متن کاملTranscription factor Sp3 knockout mice display serious cardiac malformations.
Mice lacking the zinc finger transcription factor specificity protein 3 (Sp3) die prenatally in the C57BL/6 background. To elucidate the cause of mortality we analyzed the potential role of Sp3 in embryonic heart development. Sp3 null hearts display defective looping at embryonic day 10.5 (E10.5), and at E14.5 the Sp3 null mutants have developed a range of severe cardiac malformations. In an at...
