چکیده

Tuberculosis (TB) is still a leading cause of death worldwide. Treatments remain unsatisfactory due to an incomplete understanding the underlying host–pathogen interactions during infection. In present study, weighted gene co-expression network analysis (WGCNA) was conducted identify key macrophage modules and hub genes associated with mycobacterial WGCNA performed combining our own transcriptomic results using Mycobacterium aurum-infected human monocytic macrophages (THP1) publicly accessible datasets obtained from three types infected seven different strains in various one-to-one combinations. A hierarchical clustering tree 11,533 built 198 samples, 47 distinct were revealed. We identified module, consisting 226 genes, which represented common response host infections that showed significant enrichment innate immune stimulation, bacterial pattern recognition, leukocyte chemotaxis. Moreover, by applied 74 best correlation mycobacteria infection, we top 10 hub-connecting genes: NAMPT, IRAK2, SOCS3, PTGS2, CCL20, IL1B, ZC3H12A, ABTB2, GFPT2, ELOVL7. Interestingly, apart well-known Toll-like receptor inflammation-associated other may serve as novel TB diagnosis markers potential therapeutic targets.

Weighted gene co-expression network analysis identifies specific modules and hub genes related to coronary artery disease.

BACKGROUND The analysis of the potential molecule targets of coronary artery disease (CAD) is critical for understanding the molecular mechanisms of disease. However, studies of global microarray gene co-expression analysis of CAD still remain limited. METHODS Microarray data of CAD (GSE23561) were downloaded from Gene Expression Omnibus, including peripheral blood samples from CAD patients (...

Rat Hepatocytes Weighted Gene Co-Expression Network Analysis Identifies Specific Modules and Hub Genes Related to Liver Regeneration after Partial Hepatectomy

The recovery of liver mass is mainly mediated by proliferation of hepatocytes after 2/3 partial hepatectomy (PH) in rats. Studying the gene expression profiles of hepatocytes after 2/3 PH will be helpful to investigate the molecular mechanisms of liver regeneration (LR). We report here the first application of weighted gene co-expression network analysis (WGCNA) to analyze the biological implic...

Identification of hub genes and pathways associated with retinoblastoma based on co-expression network analysis.

The objective of this paper was to identify hub genes and pathways associated with retinoblastoma using centrality analysis of the co-expression network and pathway-enrichment analysis. The co-expression network of retinoblastoma was constructed by weighted gene co-expression network analysis (WGCNA) based on differentially expressed (DE) genes, and clusters were obtained through the molecular ...

Identification of hub genes of pneumocyte senescence induced by thoracic irradiation using weighted gene co-expression network analysis

Irradiation commonly causes pneumocyte senescence, which may lead to severe fatal lung injury characterized by pulmonary dysfunction and respiratory failure. However, the molecular mechanism underlying the induction of pneumocyte senescence by irradiation remains to be elucidated. In the present study, weighted gene co‑expression network analysis (WGCNA) was used to screen for differentially ex...

Transcriptome sequencing identified hub genes for hepatocellular carcinoma by weighted-gene co-expression analysis

Hepatocellular carcinoma (HCC) is one of the most common malignancies worldwide, and it remains a challenge to understand the genetic mechanisms underlying hepatocarcinogenesis. A global gene network of differential expression profiles in HCC has yet to be fully characterized. In the present study, we performed transcriptome sequencing (mRNA and lncRNA) in liver cancer and cirrhotic tissues of ...
