چکیده

Present investigation deals with optimization of appropriate substrate concentration and incubation temperature both for growth of Saccharomyces cerevisiae and invertase production. Submerged fermentation technique was employed in the present study. The maximal production of invertase during the course of study was achieved after 48 h of incubation using initial sucrose concentration, 15.0 g l. The sugar consumption and dry cell mass were also examined. Data was subjected to kinetic analysis and on the basis of kinetic parameters such as Yp/x (amount of enzyme produced mg cell mass), Yp/s (amount of enzyme produced mg sugar consumed), Yx/s (mg cells mg substrate consumed), Ys/x (mg sugar consumed mg cell mass produced), qp (Amount of enzyme produced mg sugar consumed h), qs (mg mg cells h), qx (mg cells mg sugar consumed h), μ (mg cells produced h), it was found that temperature had a direct influence both on substrate consumption and synthesis of enzyme. Similarly, higher concentrations of sucrose in fermentation medium induced catabolite repression of yeast invertase.

Invertase Production from a Hyperproducing Saccharomyces Cerevisiae Strain Isolated from Dates

Invertase production by wild cultures of Saccharomyces cerevisiae isolated from dates available in local market is reported. Five hyperproducing yeast strains (>100 fold higher invertase activity) were kinetically analyzed for invertase production. Saccharomyces cerevisiae strain GCAII was found to be a better invertase yielding strain among all other isolates. The values of Qp and Yp/s for GCA...

Regulation of invertase synthesis by glucose in Saccharomyces cerevisiae.

Saccharomyces cerevisiae growing under repressible conditions (1% of glucose or more) produces a burst of external invertase when shifted to higher temperatures. The secretion of this invertase requires protein synthesis, but was found to be independent of RNA formation. The level of mRNA accumulated and translated was inversely proportional to the glucose present in the growth medium. These re...

Glucose sensing and signalling properties in Saccharomyces cerevisiae require the presence of at least two members of the glucose transporter family.

The kinetics of glucose transport in a number of different mutants of Saccharomyces cerevisiae with multiple deletions in the glucose transporter gene family were determined. The deletions led to differences in maximal rate and affinity for glucose uptake by the cells, dependent on the growth conditions. At the same time, there were changes in glucose repression, as determined by expression of ...

Characterization of Phosphate Membrane Transport in Saccharomyces cerevisiae CEN.PK113-5D under Low-Phosphate Conditions Using Aerobic Continuous Culture

Two different growth media, namely complex and defined media, were used to examine establishment of steady-state conditions in phosphate-limited culture system of Saccharomyces cerevisiae CEN.PK113-5D strain. Using the defined growth medium, it was possible to obtain steady state condition in the continuous culture. The effect of phosphate concentration on the growth of S. cerevisiae in pho...
