The role of tumor necrosis factor-alpha in systemic lupus erythematosus

چکیده
Murine models of systemic lupus erythematosus (SLE) have shown apparently contradictory evidence in that either (a) tumor necrosis factor (TNF) expression was low and TNF administration helpful or (b) TNF was high and TNF blockade of therapeutic benefit, depending on the mouse model investigated. In fact, TNF apparently has both effects, checking autoimmunity, at least to some degree, and fostering inflammation. TNF blockade regularly, but transiently, induces or increases autoantibodies to chromatin and to phospholipids. At the same time, open-label data suggest that TNF blockade suppresses inflammatory manifestations of SLE, and long-term benefit was seen in patients with lupus nephritis. A controlled clinical trial is under way.

Cytokines (IL-17, IL-23, and IL-33) in Systemic lupus erythematosus in Trinidad and Tobago.
Systemic lupus erythematosus (SLE) is the most common autoimmune disease. It is characterized by the presence of hundreds of autoantibodies against many organs and tissues, including the presence of a large number of autoantibodies, which are specific to self-antigens mainly of nuclear origin such as Smith antigen, double-stranded DNA (dsDNA), anti-Sjögren’s syndrome-related antigen A and B (SS...
متن کاملInfluence of 1 Alpha, 25-Dihydroxyvitamin D3 on T Helper 17 Cells and Related Cytokines in Systemic Lupus Erythematosus
Background: Systemic lupus erythematosus (SLE) is a multisystem autoimmune disease. Emerging data suggests that T helper 17 (Th17) cells play a pathogenic role in SLE and the increased number of these cells correlates with disease activity. In recent years, 1α, 25-dihydroxyvitamin D3 (1,25VitD3) has been considered as an immunomodulatory factor. Objective: To investigate the effect of 1,25VitD3...
متن کاملCytokines in systemic lupus erythematosus, London, UK
The meeting consisted of 11 talks that illustrated the complexity of the pathogenetic mechanisms underlying systemic lupus erythematosus and aimed to identify ways in which cytokine modulation might affect those mechanisms. The evidence relating to the involvement of tumour necrosis factor-alpha, interleukin-10 and BLyS in this disease was discussed in particular detail. A final discussion expl...
متن کاملCodon 72 Polymorphism of p53 Gene and Hematologic Manifestations in Patients with Systemic Lupus Erythematosus
Background: Systemic lupus erythematosus is a systemic autoimmune disorder with unclear etiology. The importance of some genes in the development of systemic lupus erythematosus has been implicated. The gene polymorphism in codon 72 has attracted a lot of attention and its role in the occurrence or progression of many cancers and autoimmune diseases especially systemic lupus erythematosus has ...

INTRODUCTION The production of tumor necrosis factor (TNF)-alpha has been deeply deregulated in systemic lupus erythematosus. We evaluated the association of -863C>A and -1031T>C polymorphisms of the TNF gene with susceptibility to and clinical manifestations of juvenile systemic lupus erythematosus. MATERIALS AND METHODS This study was performed on 70 juvenile patients with systemic lupus er...
