HLA-G proteins in cancer: do they provide tumor cells with an escape mechanism?
نویسندگان
چکیده
Convincing clinical evidence indicates that the limited success of T-cell-based immunotherapy of malignant diseases is caused, at least in part, by the ability of malignant cells to escape from immune recognition and destruction. Among the multiple escape mechanisms identified, a major role is played by changes in the expression and/or function of HLA antigens expressed by tumor cells, because they may markedly affect tumor cell-host's immune system interactions. In this article, we review the data about the aberrant expression of the nonclassical HLA class I antigen HLA-G by tumor cells. Furthermore, we discuss the possible reasons for the conflicting information in the literature about HLA-G antigen expression by malignant cells. Lastly, in light of the well-documented immunotolerant function of HLA-G, we discuss the potential role of these antigens in the escape of tumor cells from immune recognition and destruction and in the clinical course of malignant diseases.

متن کاملCorrelation of Tissue Distribution of HLA-G and HLA-E and Degree of Tumor Malignancy in Patients with Breast Cancer
Background and Aims: Human leukocyte antigen (HLA) molecules play a substantial role in T Lymphocyte-mediated adaptive immune response. Down-regulation of HLA expression may help the tumor to escape from immune surveillance. This study aimed to evaluate the correlation between HLA-G and HLA-E tissue distribution and the degree of tumor malignancy in human breast tumors. Materials and Methods:...
متن کاملAnalysis of HLA-G Gene Expression in B-Lymphocytes from Chronic Lymphocytic Leukemia Patients
The human leukocyte antigen G (HLA-G) molecule exhibits limited tissue distribution, low polymorphism and alternative splicings that generate seven HLA-G isoforms. HLA-G exerts multiple immunoregulatory functions. Recent studies indicate an ectopic up-regulation in tumor cells that may favor their escape from anti-tumor immune responses. This study it is an effort to clarify the presence of HLA...
متن کاملRole of HLA-G in tumor escape through expansion of myeloid-derived suppressor cells and cytokinic balance in favor of Th2 versus Th1/Th17.
The expression of HLA-G by malignant cells has been proposed as a tumor escape mechanism from immunosurveillance. However, although the inhibitory effect of HLA-G on antitumoral immune effectors has been documented in vitro, it remains to be resolved in vivo. In this context, the development of an animal model is now a priority to establish the proof of concept that an HLA-G(+) tumor cell devel...
متن کاملHLA-mediated tumor escape mechanisms that may impair immunotherapy clinical outcomes via T-cell activation
Although the immune system provides protection from cancer by means of immunosurveillance, which serves a major function in eliminating cancer cells, it may also lead to cancer immunoediting, molding tumor immunogenicity. Cancer cells exploit several molecular mechanisms to thwart immune-mediated death by disabling cellular components of the immune system associated with tumor recognition and r...
