چکیده

In this work we explored the structured that form when cationic liposomes are mixed with microtubules (MTs). Using synchrotron small angle X-ray diffraction (SAXRD) and transmission electron microscopy (TEM), we find two novel structures(1). For soft and intermediate rigid membranes we find that the cationic liposomes spread and coat the MTs and the external lipid layer is decorated by tubulin oligomers, forming a novel lipid-protein nanotube (LPN). By controlling the cationic lipid/tubulin stoichiometry, RCL/T, of the complex the LPN can switch between a state with open ends (Sketch 1A) to a state with closed ends with lipid caps (Sketch 1B). This form the basis for controlled drug encapsulation and release.

Cationic liposome-microtubule complexes: pathways to the formation of two-state lipid-protein nanotubes with open or closed ends.

Intermolecular interactions between charged membranes and biological polyelectrolytes, tuned by physical parameters, which include the membrane charge density and bending rigidity, the membrane spontaneous curvature, the biopolymer curvature, and the overall charge of the complex, lead to distinct structures and morphologies. The self-assembly of cationic liposome-microtubule (MT) complexes was...

Cationic membranes complexed with oppositely charged microtubules: hierarchical self-assembly leading to bio-nanotubes.

The self-assembly of microtubules and charged membranes has been studied, using x-ray diffraction and electron microscopy. Polyelectrolyte lipid complexes usually form structures templated by the lipid phase, when the polyelectrolyte curvature is much larger than the membrane spontaneous curvature. When the polyelectrolyte curvature approaches the membrane spontaneous curvature, as in microtubu...

Cell transfection in vitro and in vivo with nontoxic TAT peptide-liposome-DNA complexes.

Liposomes modified with TAT peptide (TATp-liposomes) showed fast and efficient translocation into the cell cytoplasm with subsequent migration into the perinuclear zone. TATp-liposomes containing a small quantity (<or=10 mol %) of a cationic lipid formed firm noncovalent complexes with DNA. Here, we present results demonstrating both in vitro and in vivo transfection with TATp-liposome-DNA comp...

The role of cytoskeleton networks on lipid-mediated delivery of DNA.

BACKGROUND Lipid-mediated delivery of DNA is hindered by extracellular and intracellular barriers that significantly reduce the transfection efficiency of synthetic nonviral vectors. RESULTS In this study we investigated the role of the actin and microtubule networks on the uptake and cytoplasmic transport of multicomponent cationic liposome-DNA complexes in CHO-K1 live cells by means of conf...

Evaluation of immunogenicity and protective efficacy of a liposome containing Brucella abortus S19 outer membrane protein in BALB/c mice

The use of liposome as an adjuvant and a vaccine carrier has been cited previously in the literature. It has also been shown to be effective in enhancing the immunogenicity of vaccine candidates. BALB/c mice immunized subcutaneously with outer membrane protein (OMP) of Brucella abortus S19 vaccine strain entrapped in a commercial cationic liposome (S19-OMP-liposome) for vaccine delivery, showed...
