The technique is based on the ability of the specific fluorophore PicoGreen to preferentially interact with high integrity DNA molecules, dsDNA, allowing direct fluorimetric measurement of fluorophore-dsDNA complex decay in alkaline conditions. This analytical technique allows simple and fast simultaneous analysis of DNA integrity in numerous, low DNA content samples. Research has confirmed that the Fast Micromethod is applicable for the measurement of DNA integrity in not only human biological material for medical purpose, but also in marine invertebrates for genotoxicity assessment (biomonitoring) and the estimation of harmful effects in the ecosystem. This article focuses on a comprehensive description of the method and its uses, and offers detailed descriptions and insights into samples and chemicals preparation, fluorescence analysis and calculation of DNA integrity rate. The results and features of many provided studies using this method are discussed, in order to elucidate advantages and imperfections of this analytical technique and to highlight approaches that may allow its further development.

Should sperm DNA fragmentation testing be routinely used in assessing male infertility?

Semen analysis (SA) is the cornerstone for male infertility evaluation. Variations in sperm quantity and quality may make SA an unreliable decision-making tool in providing an insight of infertility (1). Consequently, efforts have been made to upgrade techniques of male fertility analysis. In recent times, primary investigative techniques have centered around assessing sperm capacity, sperm fun...

An optimized affordable DNA-extraction method from Salmonella enterica Enteritidis for PCR experiments

In diagnostic and research bacteriology settings with budget and staff restrictions, fast and cost-effective genome extraction methods are desirable. If not inactivated properly, cellular and/or environmental DNA nucleases will degrade genomic material during the extraction stage, and therefore might give rise to incorrect results in PCR experiments. When crude cell extracts, proteinase K–treat...

DNA Damage Induced in Glioblastoma Cells by I-131: A Comparison between Experimental Data and Monte Carlo Simulation

OBJECTIVE The passage of ionizing radiation in living cells creates clusters of damaged nucleotides in DNA. In this study, DNA strand breaks induced by the beta particle of iodine-131 (I-131), have been determined experimentally and compared to Monte Carlo simulation results as a theoretical method of determining(131)I damage. MATERIALS AND METHODS In this experimental study, in order to crea...

P-18: Effects of Ethanol Consumption on Chromatin Condensation and DNA Integrity of Epididymal Spermatozoa In Rat

Background: Alcohol abuse is considered as one of the problems associated with poor semen production and sperm quality. Both acute and chronic alcohol consumption may affect spermatozoal chromatin disorders through apoptosis. Therefore, for the first time, this experimental study was performed to evaluate the effect of ethanol consumption on sperm parameters and chromatin integrity of spermatoz...

P-74: Cytochemical Evaluation of Sperm Chromatin and DNA Integrity in Couples with 3 Unexplained Recurrent Spontaneous Abortion

Background: The aim of this study was to examine the possible relationship between sperm DNA integrity and chromatin packaging evaluated by cytochemical assays, traditional sperm parameters and recurrent spontaneous abortion (RSA) of unknown origin. Materials and Methods: In this cohort study, 40 couples with a history of RSA and 40 couples with proven fertility were considered as case and cont...
