Experimental theromodynamics of the helix-random coil transition. I. Influence of polymer concentration and solvent composition in the PBG-DCA-EDC system.

چکیده
The course of the reversible helix formation of poly(7-benzyl L-glutamate) (PBG) dissolved in a mixture of dichloroacetic acid (DCA) and 1,2-dichloroethane (EDC) was followed by measuring the heat capacity and the optical rotation of the system through the transition region. The resulix of these measurements indicate that the transition enthalpy AH, the transition temperature T,, and the Zimm-Bragg parameter u depend considerably on the PBG concentration as well as on the composition of the solvent. For the standard state of infinite dilution, however, a linear extrapolation of the measured AH values results in a standard value AHo = 950 cal./mole, independent of the solvent composition. The results of the calorimetric measurements are discussed in relationship to changes in optical rotation. Some peculiarities in the measured thermodynamic and optical properties in solutions with relatively high content of dichloroacetic arid are reported.

Molecular dynamics studies on the denaturation of polyalanine in the presence of guanidinium chloride at low concentration
Molecular dynamic simulation is a powerful method that monitors all variations in the atomic level in explicit solvent. By this method we can calculate many chemical and biochemical properties of large scale biological systems. In this work all-atom molecular dynamics simulation of polyalanine (PA) was investigated in the presence of 0.224, 0.448, 0.673, 0.897 and 1.122 M of guanidinium chlorid...

We discuss the effects of the solvent composition on the helix-coil transition of a polypeptide chain. We use a simple model to demonstrate that improving the hydrogen-bonding ability of the solvent can make the transition less cooperative, without affecting the transition temperature. This effect is very different from other solvent effects which primarily influence the melting transition rath...
متن کاملCompetition for hydrogen-bond formation in the helix-coil transition and protein folding.
The problem of the helix-coil transition of biopolymers in explicit solvents, such as water, with the ability for hydrogen bonding with a solvent is addressed analytically using a suitably modified version of the Generalized Model of Polypeptide Chains. Besides the regular helix-coil transition, an additional coil-helix or reentrant transition is also found at lower temperatures. The reentrant ...
متن کاملHelix-Coil Stabilities of L-Alanine and L-Leucine in Mixed Organic Solvents
For several years, we have been engaged in evaluating the helix-coil stability constants for the naturally occurring amino acids in water.’ Specifically, we have determined the values of the parameters u and s of the Zimm-Bragg theory2 for several amino acids in aqueous solution, using the “host-guest” t e~hn ique .~ A similar series of studies to determine these same parameters in nonaqueous s...
متن کاملThe stability of helical polynucleotides: base contributions.
Estimates have been made of the main contributions of the heterocyclic bases of DNA to the free energies of the two configurations in solution, the two-stranded helix and the single-strand random coil. The magnitude and directions of the dipole moments of the adenine, thymine, guanine and cytosine base groups have been calculated by a semi-empirical molecular orbital treatment. Dipole-dipole, d...
