چکیده

The concentration of [3H]DHA was 1.0 n M . Non-specific binding was defined by the curve-fitting program and it was determined as 29% of total [”IDHA binding. The competition curve fit best to a two-site model (f.’<O.0025), with K , = 4 f 2 nM and K L = 3 f 2 ,UM ( m e a n s + s ~ ~ of three separate competition experiments which were pooled). These values were obtained by setting the affinity of [”IDHA for the two sites at 1.8 n M .

Tau isoform-specific modulation of kinesin-driven microtubule gliding rates and trajectories as determined with tau-stabilized microtubules.

We have utilized tau-assembled and tau-stabilized microtubules (MTs), in the absence of taxol, to investigate the effects of tau isoforms with three and four MT binding repeats upon kinesin-driven MT gliding. MTs were assembled in the presence of either 3-repeat tau (3R tau) or 4-repeat tau (4R tau) at tau:tubulin dimer molar ratios that approximate those found in neurons. MTs assembled with 3R...

Tau Protein and Adult Hippocampal Neurogenesis

Tau protein is a microtubule-associated protein found in the axonal compartment that stabilizes neuronal microtubules under normal physiological conditions. Tau metabolism has attracted much attention because of its role in neurodegenerative disorders called tauopathies, mainly Alzheimer disease. Here, we review recent findings suggesting that axonal outgrowth in subgranular zone during adult h...

Tauopathies, such as Alzheimer’s disease, some cases of frontotemporal dementia, corticobasal degeneration and progressive supranuclear palsy, are characterized by aggregates of the microtubule-associated protein tau, which are linked to neuronal death and disease development and can be caused by mutations in the MAPT gene. Six tau isoforms are present in the adult human brain and they differ b...

P 142: Air Pollution\'s Triggering Role in Tau Protein Hyper Phosphorylation; A Sign of Alzheimer Disease

Nowadays, air pollution is one of the major problems in developed and developing countries. In recent years, effects of air pollution on neuroinflammatory diseases such as Alzheimer disease and Parkinson disease have been studied. Researches on polluted cities citizens indicate increasing in central nervous system (CNS) inflammatory factors in comparison with clean cities; also air pollution ex...

Tau isoform expression and regulation in human cortical neurons.

Differential expression, activity, phosphorylation, and oligomerization of tau play a critical role during neuronal development and in a number of age-related neurodegenerative diseases. An experimental system that accurately models the molecular changes involved in tauopathies, particularly changes in tau isoform activity, requires the expression at physiological levels of the full complement ...
