Euglena gracilis Z ., Porphyrin Biosynthesis. Porphobilinogenase. Euglena Chloroplasts Lysed cells as well as different fractions of isolated, disrupted chloroplasts from light grown Euglena gracilis were tested for their capability of porphyrin biosynthesis. It is shown that the formation of the soluble porphobilinogenase (PBG-ase) fraction is inhibited by cycloheximide indicating its biosyntheses on 80S ribosomes, whereas the formation of the membrane bound chloroplast PBG-ase is chloramphenicol sensitive. Different pH-optima are demonstrated for the two enzymes. From the presented results it is deduced from those isolated from dark grown cells.

Dna-dependent Rna Synthesis in Chloroplasts of Euglena Gracilis
Isolated chloroplasts of Euglena gracilis have been shown to be capable of incorporating amino acids into protein (2). This incorporation is inhibited by actinomycin D, a fact which suggests that the chloroplasts might be capable of DNAdependent RNA synthesis. Such RNA synthesis is also indicated in a recent report (6) showing inhibition of chlorophyll synthesis by actinomycin. In conjunction w...
متن کاملDegeneration and regeneration of chloroplasts in Euglena gracilis grown in the presence of acetate: ultrastructural evidence.
When green cells of Euglena gracilis, strain Z, were light-grown for several months on a solid medium containing an excess of sodium acetate (1.0% instead of the normal 0.1%), some 30% of the cells were colourless. The 'acetate-bleached' organisms, isolated by plating methods and subsequently incubated in the light in a liquid medium, regained the capacity to form chlorophyll in a few days in t...
متن کاملA Study of Plastoquinones in Photochemical Reactions in the Chloroplasts of Euglena gracilis Strain Z.
Plastoquinone-9 (PQ-9) was isolated from the chloroplasts of Euglena gracilis Strain Z and spinach. The functional involvement and the structural specificity of PQ-9 in photochemical reactions was investigated in the isolated chloroplasts of Euglena gracilis. It was found that PQ-9 was required for both photoreduction of ferricyanide and photosynthetic phosphorylation in Euglena chloroplasts. T...
متن کاملUltraviolet Sensitive Factors in the Cytoplasm That Affect the Differentiation of Euglena Plastids
The bleaching phenomenon in Euglena gracilis strain z offers a unique possibility for investigating the hereditary nature of plastids. Trea tment of Euglena cultures with streptomycin, elevated temperature, or irradiation with a mild close of U V light causes the permanent loss of the capacity to develop chloroplasts in the growing cultures. Early explanations of this phenomenon were that the b...
متن کاملStudies on S-adenosylmethionine-magnesium protoporphyrin methyltransferase in Euglena gracilis strain Z.
1. An enzyme that methylates magnesium protoporphyrin was detected in extracts of light-grown and dark-grown cells of Euglena gracilis. The activity in light-grown cells is two to three times that in cells grown in the dark. 2. The activity is mainly located in the chloroplast fraction from light-grown cells and in proplastids in dark-grown cells. However, in cells grown either in the light or ...
