چکیده

A solubilized preparation from pigeon liver microsomes produced four species of phosphatidate when incubated with a mixture of linoleoyland stearoyl-CoA and sn-glycero-3phosphate. The appearance of distearoyl (15%) and dilinoleoyl (42%) species indicates that either acid may acylate both positions 1 and 2. I-Stearoyl-Z-linoleoylglycero-3-phosphate (21%) was no more abundant than l-linoleoyl-2-stearoylglycero-3-phosphate (22%), indicating that the cell-free system was not selective in esterifying these two acids. Slices of rat liver provided results in marked contrast to those above in that small amounts (1%) of disaturated phosphatidates were produced, and essentially none (3%) of the acids at position 2 was saturated. The major species produced by the slices were predominantly (75%) monoene and diene types formed by the esterification of oleate and linoleate, respectively, at position 2 with saturated acids at position 1. Choline and ethanolamine phosphoglycerides also contained disproportionately greater amounts of newly synthesized, radioactive diene species than would be expected from the endogenous abundance of these types. Endogenous species of diacylglycerophosphorylethanolamine include monoene (2%), diene (19%), tetraene (48%), and hexaene (19%), whereas the isotopic content of the newly synthesized species was approximately 14, 28, 19, and 20%, respectively.

Phospholipids in the developing soybean seed.

The distribution of phospholipids in developing soybean seeds [Glycine max (L.) Merr., var. "Chippewa 64," "Harosoy 63," "Wayne," and "Clark 63"] was followed. From 30 to 60 days after flowering expressed as mole per cent of phospholipid phosphorus phosphatidic acid decreased from 14.8 to 9.1; phosphatidylinositol increased from 0 to 9.1; phosphatidylcholine increased from 8.2 to 9.8; phosphati...

Involvement of phospholipids in triglyceride biosynthesis by developing soybean cotyledons.

The incorporation of phospholipids specifically labeled with glycerol-2(3)H and acyl-(14)C by whole cell tissues of developing soybean cotyledons (Glycine max L.) reveals that phosphatidylinositol, phosphatidylcholine, phosphatidylethanolamine, N-acylphosphatidylethanolamine, and phosphatidic acid can be metabolized to diglyceride. The diglyceride formed may be recylced into phospholipid or acy...

The interconversion of diacylglycerol and phosphatidylcholine during triacylglycerol production in microsomal preparations of developing cotyledons of safflower (Carthamus tinctorius L.).

Microsomal preparations from the developing cotyledons of safflower (Carthamus tinctorius) catalyse the acylation of sn-glycerol 3-phosphate in the presence of acyl-CoA. Under these conditions the radioactive glycerol in sn-glycerol 3-phosphate accumulates in phosphatidic acid, phosphatidylcholine, diacyl- and tri-acylglycerol. The incorporation of glycerol into phosphatidylcholine is via diacy...

Effect of freezing and cold storage on phospholipids in developing soybean cotyledons.

Freezing of plant tissue adversely affects lipid composition. Immature soybean cotyledons (Glycine max L. Merr.) var. "Harosoy 63" were frozen with liquid N(2), dry ice, or stored in a freezer (-20 C) before lipid extraction. The effects of freezing temperature, thawing rate, and cold storage on the lipid composition of frozen tissue revealed significantly higher levels of phosphatidic acid, an...
