چکیده

Studies of cell movement have many applications, such as invasion of malignant cells into healthy tissue and leukocyte migration into inflammatory sites. Cultured adherent cells usually move over their substrate in a crawling motion that varies greatly between cultures due to factors such as cell type, malignancy, mutations, confluence, substrate, or external chemical or mechanical stimuli. Cell movement can be divided into random movement (motility), and directional movement (migration). Different cell types may be characterized by their speed of movement. E.g. human fibroblasts move very slowly (12 – 60 μm/h) compared to neutrophils, which are the fastest moving leukocytes (900 – 1200 μm/h) [1].

Simultaneous two-color imaging in digital holographic microscopy

We demonstrate the use of two-color digital holographic microscopy (DHM) for imaging microbiological subjects. The use of two wavelengths significantly reduces artifacts present in the reconstructed data, allowing us to image weakly-scattering objects in close proximity to strongly-scattering objects. We demonstrate this by reconstructing the shape of the flagellum of a unicellular eukaryotic p...

Cellular Motility as a Novel Contrast Agent in Digital Holography of Tissue

Digital off-axis holography with coherence-domain depth gating is able to measure sub-cellular motion inside tumors responding to cytoskeletal anti-cancer drugs. Cellular motility is essential for virtually all cellular functions, both benign and pathological, including organelle trafficking inside the cytosol, cell division during mitosis, and cellular migration during metastasis of tumors. Mi...

Influence of salinomycin treatment on division and movement of individual cancer cells cultured in normoxia or hypoxia evaluated with time-lapse digital holographic microscopy

Most studies on new cancer drugs are based on population-derived data, where the absence of response of a small population may pass unnoticed. Thus, individual longitudinal tracking of cells is important for the future development of efficient cancer treatments. We have used digital holographic microscopy to track individual JIMT-1 human breast cancer cells and L929 mouse fibroblast cultivated ...

A Quantitative 3D Motility Analysis of Trypanosoma brucei by Use of Digital In-line Holographic Microscopy

We present a quantitative 3D analysis of the motility of the blood parasite Trypanosoma brucei. Digital in-line holographic microscopy has been used to track single cells with high temporal and spatial accuracy to obtain quantitative data on their behavior. Comparing bloodstream form and insect form trypanosomes as well as mutant and wildtype cells under varying external conditions we were able...

Measuring the deposited energy from a non-ionizing laser beam in water by digital holographic interferometry

Digital Holographic interferometry is a powerful and widely used optical technique for accurate measurement of variations in physical quantities such as density, refractive index, and etc. In this study, an experimental digital holographic interferometry setup was designed and used to measure the amount of energy changes induced by absorption of radiation from a non-ionizing infrared laser beam...
