چکیده

A weak cation exchange micro-column was incorporated into a sequential injection (SI) system to perform automatic hemoglobin (Hb) typing as an alternative way to measure HbE. Separation of HbF, HbA and HbA(2)/HbE was performed using phosphate buffer solutions in the pH range of 6-7 to create pH gradient mobile phase. The resultant chromatogram showed relative amounts of HbE to other types of hemoglobins in more quantitative detail than the conventional techniques such as dichlorophenol indophenol precipitation and micro-column anion exchange. The system is more economical than a commercially available ion-exchange HPLC analyzer for hemoglobin testing, though analysis time per run is longer due to the aspiration operation of the syringe pump of the SI system. It has been demonstrated that the system can differentiate negative (normal) from positive (HbE carriers) subjects.

A simple flow injection-reduced volume column system for hemoglobin typing.

A flow injection (FI)-reduced volume column system was developed for hemoglobin (Hb) typing to be used as an initial screening method for thalassemia. The column was packed with 140 microl diethylaminoethyl (DEAE)-Sephadex A-50 ion exchange beads. Hb can be separated using Tris-HCl buffer solution with pH gradient 8.5-6.5 and then monitored spectrophotometrically at 415 nm. The hemolysate of 40...

Evaluation of high performance liquid chromatography for routine estimation of haemoglobins A2 and F.

AIMS To compare high performance liquid chromatography (HPLC) with conventional methods for the estimation of blood haemoglobin A2 (HbA2) and haemoglobin F (HbF) concentrations in routine thalassemia screening. METHODS An HPLC system (the VARIANT Hemoglobin Testing System) was tested for precision and reproducibility in the measurement of HbA2 and HbF, and reference ranges were obtained for a...

Hemoglobin Disorders in South India

Cation exchange-high performance liquid chromatography (CE-HPLC) is increasingly being used as a first line of investigation for hemoglobinopathies and thalassemias. Together with a complete blood count, the CE-HPLC is effective in categorizing hemoglobinopathies as traits, homozygous disorders and compound heterozygous disorders. We carried out a one year study in Apollo Hospitals, Chennai (Ta...

Quantitation of hemoglobin A1a+b and hemoglobin A1c by automated "high-performance" liquid chromatography.

The glycosylated hemoglobins A1a+b and A1c have been rapidly and precisely quantitated in 5-micro L samples of human blood hemolysate (approximately 240 micrograms of hemoglobin) by cation-exchange column chromatography. Total chromatographic is 22.0 min. Proportions of Hb A1c range from 3.85 to 6.71% in normal individuals and from 4.23 to 19.90% in diabetic subjects. Within-day variation was 1...

Automated injection of uncleaned samples using a ten-port switching valve and a strong cation-exchange trap column for proteome analysis.

Nanoliter flow rate is optimum for separation in capillary column liquid chromatography coupled with tandem mass spectrometry (muHPLC-MS/MS). In order to develop a high-performance automated proteome analysis system allowing direct injection sample containing detergents, the influence of void volume varied from 0 to 5 microL on the separation performance and proteomic coverage of sample injecti...
