Kinetochore Attachment: How the Hec Can a Cell Do It?
نویسندگان
چکیده
Creating stable yet flexible attachments of spindle microtubules to the kinetochore is critical for facilitating chromosome congression, segregation, and checkpoint signaling. Two new studies have elucidated the molecular details of how the Ndc80 complex mediates this dynamic attachment.

RAMA1 is a novel kinetochore protein involved in kinetochore-microtubule attachment.
During mitosis, kinetochores need to attach to microtubules emanating from spindle poles. Several protein complexes have been shown to mediate the kinetochore-microtubule interaction. However, with the continually growing number of newly identified kinetochore proteins, it is unclear whether all major components of the kinetochore-microtubule interface have been identified. We therefore perform...
متن کاملSpindle assembly checkpoint satisfaction occurs via end-on but not lateral attachments under tension
To ensure accurate chromosome segregation, the spindle assembly checkpoint (SAC) prevents anaphase until all kinetochores attach to the spindle. What signals the SAC monitors remains unclear. We do not know the contributions of different microtubule attachment features or tension from biorientation to SAC satisfaction nor how these possible cues change during attachment. In this study, we quant...
متن کاملKinetochore function is controlled by a phospho-dependent coexpansion of inner and outer components
It is widely accepted that the kinetochore is built on CENP-A-marked centromeric chromatin in a hierarchical order from inner to outer kinetochore. Recruitment of many kinetochore proteins depends on microtubule attachment status, but it remains unclear how their assembly/disassembly is orchestrated. Applying 3D structured illumination microscopy to Xenopus laevis egg extracts, here we reveal t...
متن کاملMerotelic kinetochore attachment: causes and effects
Accurate chromosome segregation depends on the proper attachment of sister kinetochores to microtubules emanating from opposite spindle poles. Merotelic kinetochore orientation is an error in which a single kinetochore is attached to microtubules emanating from both spindle poles. Despite correction mechanisms, merotelically attached kinetochores can persist until anaphase, causing chromatids t...
متن کاملKinetochore-microtubule interactions: the means to the end.
Kinetochores are proteinaceous complexes containing dozens of components; they are assembled at centromeric DNA regions and provide the major microtubule attachment site on chromosomes during cell division. Recent studies have defined the kinetochore components comprising the direct interface with spindle microtubules, primarily through structural and functional analysis of the Ndc80 and Dam1 c...
