چکیده

The role of heme in the maturation of erythroblasts and in globin gene expression was investigated with two in vitro erythroid cell culture systems. T3-Cl2 murine erythroleukemia cells (MEIC) and primary cells from mouse fetal liver (MFL). The accumulation and/or rates of synthesis of globin protein. heme. and globin mRNA were assayed in cells in which endogenous heme, biosynthesis was reduced by the tuberculostatic drug isonicotinic acid hydrazide (INH). Under conditions in which heme synthesis was reduced 2-4-fold in dimethylsulfoxide (Me2SO) treated MELC and 5-10-fold in MFL erythroblasts, the levels of globin protein were reduced 10-fold and 2-fold. respectively. The rates of synthesis of globin protein were reduced to similar extents without a concomitant decrease in total protein synthesis. The levels of globin mRNA in INH-treated MELC and MFL cells were 2-7-fold lower than those in untreated cells. In MELC cells cultured with both INH and hemin. the globin protein and mRNA levels were the same as in cells cultured with hemin alone. The globin mRNA content of cells cultured with Me2SO plus INH was low but increased within 5 hr after the addition of hemin. Thus. hemin overcame the inhibitory effects of INH. These data are consistent with the hypothesis that heme affects globin gene expression at the pretranslational, as well as the translational level.

The effect of water borne nickel on iron metabolism and heme biosynthesis in common carp (Cyprinus carpio)

Nickel is an essential element for all living organisms such as microorganisms, plants and animals. When nickel concentration exceeds the necessary concentration, could be toxic, and likewise causes adverse effects in living organisms. In this study, following determining nickel LC50-96h for common carp (Cyprinus carpio), nickel sub-lethal treatments including 0 (control), 0.055, 0.275, 0.572, ...

Inhibition of Heme Polymerization, the Mechanism of Antimalarial Activity in Phlomis caucasica Rech.f. (Lamiaceae)

Background: Malaria is one of the most important parasitic diseases in the world caused by Plasmodium species. The malaria parasite digests hemoglobin in vacuole to amino acids and heme. Plasmodium has got several detoxification mechanisms to protect itself from toxic heme. The most important mechanism is heme polymerization. Identifying compounds that inhibit heme polymerization is an approach...

Inhibition test of heme detoxification (ITHD) as an approach for detecting antimalarial agents in medicinal plants

Background and objectives: There are several methods to assess the in vitro capability of heme inhibitory activity of antimalarial compounds; most of them require some specific equipment or toxic substances and sometimes the needed materials are not accessible. Regarding the necessity and importance of optimizing and standardizing experimental conditions, the present s...

delta-Aminolevulinic acid synthetase in erythroblasts of patients with pyridoxine-responsive anemia. Hypercatabolism caused by the increased susceptibility to the controlling protease.

Properties of delta-aminolevulinic acid synthetase in erythroblasts of patients with pyridoxine-responsive anemia were investigated with special reference to the protease in mitochondria of erythroblasts. delta-Aminolevulinic acid synthetase activity in erythroblasts of patients with this disease before treatment was extremely decreased, whereas it gradually increased in parallel with the impro...
