Origin of the Electron Microscope.

چکیده

In 1878, Ernst Abbe (1) proved that the resolution of the optical microscope is limited by the wavelength of light. This meant that even when immersion optics and ultraviolet light are used, the smallest detail that can ever be resolved optically is of the order of 100 millimicrons, or 10 angstroms. No means were conceived of resolving finer detail until two discoveries were made: (i) the wave properties of the moving electron, postulated by de Broglie, on theoretical grounds, in 1924 (2) and verified by Davisson and Germer (3) and by Thomson and Reid (4) in 1927; and (ii) the discovery by Busch (5) in 1926-1927 of the analogy between the effect of a magnetic coilthe focusing coil used since 1899 (6)on an electron beam and the effect of a convex lens on a light beam. Since the wavelength of the moving electron is smaller by many orders of magnitude than the shortest wavelength of light, these discoveries made it conceivable that extremely small objects might be imaged with an electron beam and electron lenses.

origins rand historical development of the electron microscope

The history of the electron microscope is traced from the earliest speculations on the possibility of a new type of microscope, whose performance would not be limited by the wavelength of light, to its successful realization in the form of an electron microscope that could be made readily available to physicists and biologists. The historical development did not follow the smooth, predictable c...

Light and electron microscope of grass carp (Ctenopharyngodon idella) organs following exposure to various sublethal concentrations of diazinon

Histopathological effects of different sublethal concentrations (1, 2 and 4 mg/l) of diazinon, an organophosphorus pesticide, on gill, liver, spleen and kidney of grass carp (Ctenopharyngodon idella) were studied after 1, 7, 15, 30 and 45 days post-exposing fishes weighting 850 ± 155 g to the toxicant for 12 hours at 18-22ºC. In addition, tissues of nostril were examined for transmission electr...

Environmental scanning electron microscopy for biology and polymer science

Electron microscopy of materials in a gaseous environment has its origins as far back as about the 1950s for the transmission electron microscope (TEM), later followed by developments for the scanning electron microscope (SEM). Focusing on the latter, this review outlines the history and commercial development of the technology, culminating in the environmental SEM (ESEM), and the techniques fo...

Green Synthesis of Zinc Oxide Nanoparticles using Garlic skin extract and Its Characterization

Plant-mediated synthesis of metal oxide nanoparticles is a promising alternative to the traditional method of physical and chemical synthesis. In this paper, we report the green synthesis of zinc oxide nanoparticles (ZnONPs) by a biological method. During the study, Zinc oxide nanoparticles were synthesized by Allium sativum skin (garlic skin) extract. Formation of zinc oxide nanoparticles has ...

Effects of Short-Term Exposure to Sublethal Concentrations of Silver Nanoparticles on Histopathology and Electron Microscope Ultrastructure of Zebrafish (Danio Rerio) Gills

Background: The increasing use of nanomaterials and nanoproducts has increased the possibility of contamination of the environment, which may have adverse effects on different organisms. The aim of this study was to evaluate the effects of silver nanoparticles on histopathology and gill ultrastructure of zebrafish (Danio rerio) under laboratory conditions. Methods: Zebrafish were exposed to ...
