J. Physiol. 275 (Heart Circ. Physiol. 44): H1957–H1962, 1998.—To study the effects of mechanical constraints on the Ca21 affinity of cardiac troponin C, we analyzed the tension and aequorin light (AL) responses to sinusoidal length changes (5–10% of the initial muscle length) in aequorin-injected, tetanized cardiac muscles. The amplitude of the quasi-sinusoidal tension and AL responses decreased with increasing lengthperturbation frequency from 0.5 to 1 Hz at 24°C and from 1 to 3 Hz at 30°C. The increase in AL corresponded well to the decrease in tension; likewise, the decrease in AL to the increase in tension and the tension response lagged behind the length change. A further increase in frequency (.1 Hz at 24°C and .3 Hz at 30°C) markedly increased the amplitude of the tension responses but decreased the amplitude of the AL responses. The increase in AL lagged behind the decrease in tension; likewise, the decrease in AL lagged behind the increase in tension, and the tension response led the length change. From previous mechanistic interpretations of the frequency dependence of the amplitude of tension response, we argue that the Ca21 affinity of cardiac troponin C changes in parallel with the active tension (i.e., the number of active cross bridges) but not with the passive tension produced by the length perturbation-induced cross-bridge strain.

Dynamic relations among length, tension, and intracellular Ca2+ in activated ferret papillary muscles.

Responsiveness in Aequorin-Loaded Ferret Myocardium

The influence of endothelin 1 on intracellular Ca2' transients and isometric contractions was investigated in ferret papillary muscles loaded with the Ca2+-regulated bioluminescent indicator aequorin. In concentrations of 3 X 10-9 to 1x 10` M, endothelin produced dose-dependent increases in the amplitudes of both aequorin light signals (maximum, 31±12%) and developed tension (maximum, 64±13%). ...

Physiol. 277 (Heart Circ. The aequorin method...

Maximal Ca-Activated Force Elicited by Tetanization of Ferret Papillary Muscle and Whole Heart: Mechanism and Characteristics of Steady Contractile Activation in Intact Myocardium

Rapid (8-12 Hz) stimulation of intact heart muscle treated with ryanodine results in steady contractile activation known as tetanus, the amplitude of which can be graded by changing extracellular Ca concentration ([Ca]0). The mechanism of the sustained force generation was explored in ferret papillary muscles by measuring membrane potential and by determining the responsiveness of force and int...

Endothelin Reverses the Effects of Acidosis on the Intracellular Ca2' Transient and Contractilit in Ferret Myocardium

Endothelin may play an important role in modulating myocardial contractility under certain pathophysiological conditions. To determine whether endothelin beneficially modulates myocardial contractility in the common clinical condition of acidosis, we compared the effects of endothelin-1 on intracellular Ca2' transients and isometric contractions under normal (extracellular pH [pH.] 7.4) and aci...
