Leukemia-associated Antigens Detected by a Nonhuman Primate Antiserum to a Ph’+ Myelogenous Leukemia Cell Line

چکیده
Blood, Vol. 50, No. 1 (July), 1977 115 The specificity of an antiserum (titer 1:500) raised in a monkey against K-562 undifferentiated cells with the Philadelphia chromosome (Ph’) was determined. Antibody-dependent, complement-mediated cytotoxicity was demonstrated for both bone marrow cells and peripheral leukocytes from patients with myelogenous and lymphatic leukemias, but not for mature normal hematopoietic cells and those from patients with leukemia in remission. A monospecific antiserum for K-562 cells and peripheral Ieukocytes from three patients with chronic myelogenous leukemia (CML) was obtained after multiple absorptions with peripheral Ieukocytes from patients with acute and chronic lymphatic leukemias as well as acute myelogenous leukemia. The minimal number of Ph’ + myelogeneous leukemia cells needed to remove the antibody completely from 1 ml of the immune serum was found to be 12 x i0 K-562 cells versus 25 x 1 peripheral Ieukocytes (52% immature and 48% mature) in different stages of maturation from a patient with CML. Other results indicated that progenitors of normal and undifferentiated malignant cells of hematopoietic origin might share some common antigenic determinants that reacted with this antiserum but that could be removed by appropriate absorptions.

Nonhuman primate antisera defining leukemia-associ ated and normal Tand B-lymphocyte-associated anti gens were used to characterize serologically the cells from acute myelomonocytic leukemia (AMML) and chronic myelogenous leukemia patients in blast crisis (CML-BC). Earlier studies demonstrated that nonhuman primate an tisera to chronic lymphocytic leukemia, acute myelogen ous leukemia, and chro...

Lymphocyte-dependent antibodies (LDA's) directed against antigenic determinants present on lymphoblastoid cell lines as well as human leukemia blast cells were demonstrated in heterologous antisera obtained by immunizing rabbits with a membrane fraction from RPMI-4265 (a lymphoblastoid cell line derived from a patient with chronic myelogenous leukemia). LDA was present at high titers against B-...

متن کاملFunction of Neutrophils in Different Phases of Chronic Myelogenous Leukemia
In chronic myelogenous leukemia (CML), the mature granulocytes originate from a stem cell line harboring an abnormal chromosome, therefore it is possible that metabolic-functional abnormalities occur in the morphologically mature cells. In the present study, the phagocytic activity including intracellular killing, nitro blue tetrazolium (NBT) reduction, and phagocytosis were studied in 37 CML p...
