Polymeric Nanoparticles and Cancer: Lessons Learnt from CRLX101
نویسندگان
چکیده
CRLX101 (formerly IT-101) is a novel cyclodextrin-based polymer that selfassembles into nanoparticles. CRLX101 was designed to deliver sustained levels of the cytotoxic agent camptothecin (CPT) to tumour cells while minimizing delivery to normal tissue. Covalent conjugation of hydrophobic CPT to the cyclodextrin polymer increases its water solubility by three orders of magnitude and prevents inactivation through spontaneous lactone ring opening, a process that occurs rapidly at physiologic pH. CPT derivatives such as irinotecan (CPT-11, Camptosar; Pfizer) and topotecan (TPT, Hycamtin; GlaxoSmithKline) demonstrate clinical utility for the treatment of advanced solid tumours. The primary cellular target of

Correlating preclinical animal studies and human clinical trials of a multifunctional, polymeric nanoparticle.
Nanoparticles are currently being investigated in a number of human clinical trials. As information on how nanoparticles function in humans is difficult to obtain, animal studies that can be correlative to human behavior are needed to provide guidance for human clinical trials. Here, we report correlative studies on animals and humans for CRLX101, a 20- to 30-nm-diameter, multifunctional, polym...
متن کاملCRLX101 (formerly IT-101)–A Novel Nanopharmaceutical of Camptothecin in Clinical Development
CRLX101 (formerly IT-101) is a first-in-class nanopharmaceutical, currently in Phase 2a development, which has been developed by covalently conjugating camptothecin (CPT) to a linear, cyclodextrin-polyethylene glycol (CD-PEG) co-polymer that self-assembles into nanoparticles. As a nanometer-scale drug carrier system, the cyclodextrin polymeric nanoparticle technology, referred to as "CDP", has ...
متن کاملQuantum chemical study of Interaction of PLGA polymeric nanoparticles as drug delivery with anti-cancer agents of thiazoline
Thiazoles derivatives are consisted in chemical compounds such as antimicrobial and anticancer medicine. Since polylactic-co-glycolic acid (PLGA) polymeric nanoparticles has been conversed about nanomedicine applications and particularly as drug delivery systems. Because of molecular self-assemblies and biodegradability of PLGA polymer, it can be used to carry anti-cancer and antimicrobial drug...
متن کاملCancer Therapy: Preclinical Translational Impact of Nanoparticle–Drug Conjugate CRLX101 with or without Bevacizumab in Advanced Ovarian Cancer
Purpose: Increased tumor hypoxia and hence elevated hypoxia-inducible factor-1a (HIF1a) is thought to limit the efficacy of vascular endothelial growth factor (VEGF) pathway–targeting drugs by upregulating adaptive resistance genes. One strategy to counteract this is to combine antiangiogenic drugs with agents able to suppress HIF1a. One such possibility is the investigational drug CRLX101, a n...
متن کاملCRLX101 nanoparticles localize in human tumors and not in adjacent, nonneoplastic tissue after intravenous dosing.
Nanoparticle-based therapeutics are being used to treat patients with solid tumors. Whereas nanoparticles have been shown to preferentially accumulate in solid tumors of animal models, there is little evidence to prove that intact nanoparticles localize to solid tumors of humans when systemically administered. Here, tumor and adjacent, nonneoplastic tissue biopsies are obtained through endoscop...
