The Angiogenic Sprouting of Endothelial Cells in Three-dimensional Collagen Gel Matrix
نویسندگان
چکیده
It is well known that angiogenesis is an essential biological event. To understand fully how the blood vessel grows and functions in the angiogenic process, we should consider the construction of in vivo-like environment, such as threedimensional (3D) extracellular matrix (ECM). However, most of in vitro studies of the angiogenesis have been relied on two-dimensional (2D) culture or 3D Matrigel culture platforms, which do not account for the in vivo situations. To overcome these limitations in the studies of the angiogenesis, we used a simple microfluidic method incorporating the 3D ECM into the surrounding endothelial cells (ECs) channel.

Tensional forces in fibrillar extracellular matrices control directional capillary sprouting.
During angiogenesis, anastomosing capillary sprouts align to form complex three-dimensional networks of new blood vessels. Using an endothelial cell spheroid model that was developed to study endothelial cell differentiation processes, we have devised a novel collagen gel-based three-dimensional in vitro angiogenesis assay. In this assay, cell number-defined, gel-embedded endothelial cell spher...

Background: Norepinephrine plays a trophic role in the control of cell replication and differentiation in target cells that express adrenergic receptors. Methods: In this study, we have tested the influence of infraphysiological, physiological and supraphysiological concentrations (0.0001 nM, 1 nM, 10000 nM) of human norepinephrine on the proliferation of breast cancer cells (human breast adeno...
متن کاملThree-Dimensional Characterization of Mechanical Interactions between Endothelial Cells and Extracellular Matrix during Angiogenic Sprouting.
We studied the three-dimensional cell-extracellular matrix interactions of endothelial cells that form multicellular structures called sprouts. We analyzed the data collected in-situ from angiogenic sprouting experiments and identified the differentiated interaction behavior exhibited by the tip and stalk cells. Moreover, our analysis of the tip cell lamellipodia revealed the diversity in their...
متن کاملAngiogenesis Following Three-Dimensional Culture of Isolated Human Endometrial Stromal Cells
Background Endometriosis is the presence of endometrial tissue outside of the uterine cavity and is the most common gynecologic disorder in women of reproductive age. We have preliminary evidence that in the presence of a 3-dimensional (3-D) fibrin matrix, human endometrial glands, stroma, and neovascularization can develop in vitro, mimicking the earliest stages of endometriosis. The aim of th...
متن کاملHuman Coagulated Plasma as a Natural and Low Cost Matrix for in vitro Angiogenesis
Background: Angiogenesis, the development of new blood vessels, is an important process in tissue development and wound healing, but becomes pathologic when associated with solid tumor growth, proliferative retinopathies, and rheumatoid arthritis. Accurate and reliable qualification of neovascular (angiogenic) response, both in vitro and in vivo, is an essential requirement for the study of new...
