چکیده

All cells, inc luding h u m a n hemopoietic cells and lines derived from them, possess numerous surface proteins. In most h u m a n leukocytes and lymphoid cell lines several dozen proteins can be detected in membrane preparations, but so far only those proteins encoded by the various HLA loci have been extensively characterized. Vir tual ly noth ing is known about the great majori ty of lymphocyte cell surface proteins. In this paper, we provide pre l iminary characterization of a previously undescribed ant igen that is p rominent on the surface of all d ividing blood cells examined and describe its dis t r ibut ion on a variety of normal and transformed h u m a n cells.

Novel surface antigen expressed on dividing cells but absent from nondividing cells

Radioimmunoprecipitation followed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis was used to study the distribution on human lymphoid cells of a previously undescribed surface antigen recognized by several heteroantisera. A glycoprotein with a 90,000 mol wt (under reducing conditions) was detected on all cell lines tested including T, B, null, and myeloid cell lines, although the ...

Effect of Heavy Metals on Silencing of Engineered Long Interspersed Element-1 Retrotransposon in Nondividing Neuroblastoma Cell Line

Background: L1 retrotransposons are the most active mobile DNA elements in human genome. Unregulated L1 retrotransposition may have deleterious effect by disrupting vital genes and inducing genomic instabilities. Therefore, human cells control L1 elements by silencing their activities through epigenetic mechanisms. It has been shown that cell division and heavy metals stimulate the frequency of...

Cyclophilin A-dependent restriction of human immunodeficiency virus type 1 capsid mutants for infection of nondividing cells.

Among retroviruses, lentiviruses are unusual in their ability to efficiently infect both dividing and nondividing cells, such as activated T cells and macrophages, respectively. Recent studies implicate the viral capsid protein (CA) as a key determinant of cell-cycle-independent infection by human immunodeficiency virus type 1 (HIV-1). We investigated the effects of the host cell protein cyclop...

A CRISPR/Cas9 approach reveals that the polymerase activity of DNA polymerase β is dispensable for HIV-1 infection in dividing and nondividing cells

Retrovirus integration into the host genome relies on several host enzymes, potentially including DNA polymerase β (Pol β). However, whether human Pol β is essential for lentivirus replication in human cells is unclear. Here, we abolished DNA polymerase β (Pol β) expression by targeting its DNA polymerase domain with CRISPR/Cas9 in human monocytic THP-1 cells to investigate the role of Pol β in...

The kinetics of T cell antigen receptor expression by subgroups of CD4+8+ thymocytes: delineation of CD4+8+3(2+) thymocytes as post- selection intermediates leading to mature T cells

Cortical thymocytes from adult mice, separated on the basis of coexpression of CD4 and CD8 or of binding of high levels of peanut agglutinin (PNA), were subdivided according to the level of expression of the T cell receptor (TCR)-CD3 complex. The incidence of dividing cells in the resultant subpopulations was determined by DNA staining. Precursor-product relationships and the timing of TCR-CD3 ...
