چکیده

Previous studies have shown that Rhodotorula gracilis is capable of forming large quantities of fat under suitable conditions (Enebo et al., 1946; Pan et al., 1949; Steinberg and Ordal, 1954a). It also has been demonstrated that growth and fat formation takes place in two distinct phases (Enebo et al., 1946). Steinberg and Ordal (1954a, 1954b) have studied the fattening phase in some detail; however, very little is known about the optimum conditions for growth of this organism. There is, in general, a lack of accurate published in formation on the effects of pH, temperature, and aeration on the growth of yeasts. Only a few studies have appeared in the literature describing the use of pH controlling equipment in fermentation studies (Finn et al., 1950; Kempe et al., 1950; Neish and Blackwood, 1951; Maxon and Johnson, 1953; Lakata, 1954; Steinberg and Ordal, 1954a; McKee, 1955). In most cases only the initial pH was adjusted and observed and, where temperature or other conditions were variable, generally no pH control was used. There are no temperature studies reported in the literature for R. gracilis. Enebo et al. (1946) used a temperature of 28 C for both growth and fat formation. Every known investigator since that time has used this temperature. Several investigators have made quantitative studies on the effect of aeration on yeast growth (Olson and Johnson, 1949; White and Munns, 1951b; Maxon and Johnson, 1953). The only study on the effect of aeration on R. gracilis was made by Pan et al. (1949) who investigated the effect of air flow rate on combined growth and fattening. An increase of air flow rate from 0.1 to 1 volume of air per minute had no effect on the efficiency of sugar utilization nor on fat content. No ineasure of the effective oxygen-absorption rate was given. Agarwala and Peterson (1950) and Chiao and Peterson (1953) reported on the efficiency of sugar utilization by R. gracilis. However, the medium used by these workers contained nonsugar carbon compounds which could be utilized by the organism, which makes any interpretation of their results equivocal. The object of this investigation was to determine the effect of pH and temperature on total amount and rate of growth of R. gracilis under conditions of controlled

Effect of macro green algae extract on tomato (Lycopersicum sculentum Mill.) seedling growth characteristics

The effect of macro green algae (Chaetomorpha gracilis) inf Bandar-Abbas and filamentous green algae (Spirogyra sp.) in the Sirch river (Kerman Province) were assayed on growth characteristics of the tomato (Lycopersicum sculentum). In the seed priming experiment, we collected hot water extracts of green algae (Chaetomorpha gracilis) from Bandar Abbas seashores and filamentous green algae (Spir...

Chemical mechanism of D-amino acid oxidase from Rhodotorula gracilis: pH dependence of kinetic parameters.

The variation of kinetic parameters of d-amino acid oxidase from Rhodotorula gracilis with pH was used to gain information about the chemical mechanism of the oxidation of D-amino acids catalysed by this flavoenzyme. d-Alanine was the substrate used. The pH dependence of Vmax and Vmax/Km for alanine as substrate showed that a group with a pK value of 6.26-7.95 (pK1) must be unprotonated and a g...

Kinetic mechanism of D-amino acid oxidases from Rhodotorula gracilis and Trigonopsis variabilis.

The reaction of two D-amino acid oxidases from the yeasts Rhodotorula gracilis and Trigonopsis variabilis with the substrates alanine and valine in their 2-1H and 2-2H forms was studied employing the stopped-flow spectrophotometric technique. The turnover numbers at infinite substrate and oxygen concentrations were: 20,700/4,250 and 1,730/360 ([2-1H]/[2-2H]alanine and valine, respectively) for ...

FAD ANALOGUES AS ACTIVE SITE PROBES OF Rhodotorula gracilis D-AMINO ACID OXIDASE

The wide range of chemical reactivity exhibited by flavoprotein enzymes is determined fundamentally by the nature of the protein moiety. Studies on the interactions of the isoalloxazine nucleus of flavin with the enzyme are therefore ~ important in understanding this reactivitYi flavin analogues are extremely useful probes for elucidating structure-function relationships (1, 2). We have used a ...

Studies on the Reaction Mechanism of Rhodotorula gracilis

We have studied D-amino-acid oxidase from Rhodotorula gracilis by site-directed mutagenesis for the purpose of determining the presence or absence of residues having a possible role in acid/base catalysis. Tyr-223, one of the very few conserved residues among D-aminoacid oxidases, has been mutated to phenylalanine and to serine. Both mutants are active catalysts in turnover with D-alanine, and ...
