چکیده

UMEROUS thymineless mutants of E. coli, Salmonella, and other bacteria N can be obtained by the aminopterin and thymine method (AM-T method) ( OKADA, YANAGISAWA and RYAN 1961 ; OKADA, HOMMA and SONOHARA 1962). However, in order to sustain normal growth, all the thymineless mutants thus obtained require ten to 20 times the concentration of thymine required by E. coli 15 T(COHEN and BARNER 1954). Similar observations were also reported by BREITMAN and BRADFORD (1964) and by HARRISON (1965). Since a number of mutants with a low thymine requirement, similar to E. coli 15 T-, have been spontaneously obtained from the high thymine requirers, it is very likely that low ihymine requirers are derived from high as a result of a second mutation. Thus, if thc genes concerned with the first and second mutations were designated as thy and thy-R respectively, the mutational sequences may be expressed:

Mutational Screening in Exon 6 of the PSEN2 Gene in Iranian Patients with Late-Onset Alzheimer\'s Disease

Background and Aims: One of the most important genes involved in Alzheimer's disease (AD) is the presenilin2 (PSEN2) gene, which is one of the main constituents of the gamma-secretase complex. Mutations in this gene promote the formation of amyloid plaques resulting in AD. The study aimed to evaluate the mutation variant in exon 6 of the PSEN2 gene in patients with Late-Onset AD (LOAD). Due to ...

Mutational analysis of ARSB gene in mucopolysaccharidosis type VI: identification of three novel mutations in Iranian patients

Objective(s): Mucopolysaccharidosis VI (MPS VI) or Maroteaux-Lamy syndrome is a rare metabolic disorder, resulting from the deficient activity of the lysosomal enzyme arylsulfatase B (ARSB).  The enzymatic defect of ARSB leads to progressive lysosomal storage disorder and accumulation of glycosaminoglycan (GAG) dermatan sulfate (DS), which causes harmful effects on various organs and tissues an...

Separating substrate recognition from base hydrolysis in human thymine DNA glycosylase by mutational analysis.

Human thymine DNA glycosylase (TDG) was discovered as an enzyme that can initiate base excision repair at sites of 5-methylcytosine- or cytosine deamination in DNA by its ability to release thymine or uracil from G.T and G.U mismatches. Crystal structure analysis of an Escherichia coli homologue identified conserved amino acid residues that are critical for its substrate recognition/interaction...
