The A protein of the tryptophan synthetase of Escherichia coli. Purification, crystallization, and composition studies.

چکیده
Previous studies (2, 4) have led to the conclusion that Reaction 3 is probably the physiologically essential reaction in the biosynthesis of tryptophan in E. coli. Studies of the relationship between gene structure and protein structure which are presently being carried out with the A component of this system (5-8) have necessitated investigations of the properties, composition and primary structure of the A protein. This communication describes the isolation, crystallization, amino acid composition, and several physical and chemical properties of the A protein of the tryptophan synthetase of E. coli.

Cloning and evaluation of gene expression and purification of gene encoding recombinant protein containing binding subunit of coli surface antigens CS1 and CS2 from Enterotoxigenic Escherichia coli
Background & Objective: Enterotoxigenic Escherichia coli (ETEC) is a major causative agent of diarrhea. Enterotoxins and the colonization factors (CFs) are major virulence factors in ETEC infections. The bacterium binds to the intestinal epithelial cell surface through colonization factors and produces enterotoxins that cause excessive fluid and electrolyte secretion in the lumen of the intesti...
متن کاملAnthranilate synthetase. Partial purification and some kinetic studies on the enzyme from Escherichia coli.
Anthranilate synthetase was purified by ammonium sulfate precipitation and gel filtration from extracts of an episomebearing Escherichia coli mutant grown under conditions of tryptophan pathway derepression. This purification represented an l&fold increase in specific activity over the activity of the derepressedmutant extract. Starch gel electrophoresis and ultracentrifugation revealed only sl...
متن کاملProduction, Purification and Characterization of Chicken Egg Yolk Monoclonal Antibody Against Colonization factor antigen -1 of Enterotoxigenic Escherichia coli Causing Diarrhea
Enterotoxigenic Escherichia coli (ETEC) causes diarrhea in both humans and animals. The contaminated food and water are the most common vehicles for ETEC infection. The colonization factor antigen (CFA-1) is a fimbriae protein that promotes adherence of the ETEC strain to the epithelium of the small intestine of the host. In this study IgY proteins were produced against the CFA-1 of ETEC in imm...
متن کاملTryptophan Operon of Escherichia coli: Purification and Characterization of Component I
A procedure employed in the purification of anthranilate synthetase component I of Escherichia coli is described. The purified component appears homogeneous by starch gel electrophoresis and by sedimentation analysis. A molecular weight of 60,000 was estimated by gel filtration on Sephadex G-100. This value is consistent with the molecular weight estimated from the sedimentation and diffusion c...
متن کاملThe amino acid sequence of the A protein (alpha subunit) of the tryptophan synthetase of Escherichia coli.
Tryptic digestion of the A protein (a! subunit) of the tryptophan synthetase of Escherichia coli and separation of tryptic peptides by column and paper chromatography yielded 22 nonoverlapping peptides of the total of 25 anticipated on the basis of total amino acid composition of the A protein. The amino acid composition and sequence of each purified peptide were determined. These peptides acco...
