Anionic nucleotide--lipids for in vitro DNA transfection.

چکیده
A family of new anionic nucleotide based lipids featuring thymidine-3'-monophosphate as nucleotide and 1,2-diacyl-sn-glycerol as lipid moiety for in vitro delivery of nucleic acids is described. The nucleotide lipids were prepared in three steps starting from 1,2-diacyl-sn-glycerols and 2'-deoxythymidine-3'-phosphoramidite. Gel electrophoresis experiments show that nucleotide-based lipid-DNA complexes are observed at Ca(2+) concentration higher than 1 mM. The transfection experiments carried out on mammalian Hek cell lines clearly demonstrate that the nucleotide moiety enhances the transfection efficacy of the natural anionic DPPA and DPPG lipids. SAXS studies indicate that the enhancement in transfection for nucleotide-based lipid formulations compared to those of the abasic natural derivative (DPPA) is likely due to the presence of the 2D columnar inverted hexagonal phase (H(II)) with a unit cell parameter a = 69.1 A in the nucleotide lipid formulations. The cytotoxicity studies of lipoplexes, evaluated against Hek cells using an MTS assay, revealed that palmitoyl nucleotide derivative complexes were not toxic even after 4 h of incubation, thus indicating that the anionic nucleotide lipids presented in this work offer an alternative to cationic transfection reagents.

Synthesis and properties of novel tetraalkyl cationic lipids.
The synthesis, physical properties, and transfection potencies of two representives of a new class of divalent, tetraalkyl cationic lipids is described. These cationic lipids are dimers of N,N-Dioleyl-N,N-dimethylammonium chloride (DODAC) joined by a hydrocarbon tether three or six carbons in length (TODMAC3 and TODMAC6, respectively). It is shown that TODMAC6 can display improved transfection ...
متن کاملThe antibiotic and DNA-transfecting peptide LAH4 selectively associates with, and disorders, anionic lipids in mixed membranes.
The histidine-rich amphipathic peptide LAH4 has antibiotic and DNA delivery capabilities. The peptide has a strong affinity for anionic lipids found in the outer membrane of bacterial membranes. A role for anionic lipids in release of cationic plasmid-containing complexes has been proposed previously, and disruption of membrane asymmetry and presentation of phosphatidylserine (PS) in the membra...

The development of efficient and safe carrier system to transfer DNA into cells is essential in non-viral gene therapy. The aim of the present study was to evaluate the effect of linear polyetheneimine (lPEI) (2500 Da) on the physicochemical and biological properties of lipopolyplexes constructed from liposomes and lPEI. Materials and Methods Different lipopolymers were synthesized from lPEI ...
متن کاملCalcium Effect on Anionic Bicelle-DNA Complexes
Different from The liposome-DNA complexes, which were well-known to be used as nonviral vectors for improving the transfection efficiency of gene delivery, we demonstrate a novel type of anionic lipid-DNA (AB-DNA) complexes based on using typical disk-shaped bicelles. Having the advantage of lower cytotoxicity, the approach was proposed to induce anionic DPPG doped bicelles for forming the lame...
متن کاملFormation of divalent ion mediated anionic disc bicelle-DNA complexes.
Disc-shaped bicelles are formed by mixing long-chain lipids with short-chain lipids at suitable molar ratios and they have a relatively uniform size, typically around a few tens of nanometers in diameter. Different from the typically formulated cationic or anionic liposome–DNA complexes, which are used as nonviral vectors for improving the transfection efficiency of gene therapy, a novel way of...
