Donnellan, J. Edward, Jr. (U.S. Army Natick Laboratories, Natick, Mass.), Ella H. Nags, and Hillel S. Levinson. Chemically defined, synthetic media for sporulation and for germination and growth of Bacillus subtilis. J. Bacteriol. 87:332-336. 1964.-From 90 to 130 mg (dry weight) of spores (about 1% dark forms) were obtained (per liter) from a chemically defined, synthetic medium, with a two-phase (polyethylene glycol-potassium phosphate) harvest procedure. Optimal sporulation occurred when glucose and glutamic acid were at a concentration of 10 mm in the medium. Ca(++) and Mn(++) were required for sporulation. Heat resistance, dipicolinic acid content, and properties of germination and postgerminative development of spores grown in different concentrations of Ca(++) were investigated. Heat shock did not increase germination of spores derived from the synthetic medium. A synthetic medium, in which spore germination, emergence, and first cell division approached synchrony, was devised.

Single, chemically defined sporulation medium for Bacillus subtilis: growth, sporulation, and extracellular protease production.
The composition and application of a single, chemically defined medium or growth and sporulation of Bacillus subtilis is described. At 37 degrees C cells grew with a doubling time of about 40 min; cultures attained near-maximal spore formation (70 to 80% by 12 h after the end of exponential growth and produced 1 X 10(9) to 2 X 10(9) heat-resistant free spores at 24 h. Dipicolinic acid productio...
متن کاملConditional dihydrostreptomycin resistance in Bacillus subtilis.
Mutants resistant to dihydrostreptomycin were isolated and genetically analyzed in Bacillus subtilis. Two new classes of mutants distinct from the ribosomal strA locus were found. One class, strB, was located between metC3 and ura-1 on the chromosome. The second class, strC, mapped in the spore gene region close to the spoA locus. Both mutant classes were resistant to dihydrostreptomycin during...
متن کاملEnhanced Spore Production of Bacillus subtilis Grown in a Chemically Defined Medium
Spores of Bacillus subtilis are being used as probiotics and competitive exclusion agents for animal consumption. Commercial production media often include relatively expensive components of animal origin that are a potential source for the presence of adventious agents, therefore undesirable for use in production scale. In this study a new animal-free component, chemically defined medium, was ...
متن کاملSpontaneous transformation and its use for genetic mapping in Bacillus subtilis.
Using a simple semi-synthetic competence and sporulation medium (CSM), we found evidence that Bacillus subtilis cells transformed in the competence phase can sporulate, indicating that genetic information acquired during the competence phase is inherited by the next generation after germination of the transformed spores. Moreover, the results from mixed cell culture experiments suggest that spo...
متن کاملBacillus subtilis deoxyribonuclease activity specific for single-stranded deoxyribonucleic acid: cellular site and variations during germination and sporulation.
The endonuclease of Bacillus subtilis specific for single-stranded deoxyribonucleic acid is absent in spores, appears during germination only after the start of deoxyribonucleic acid synthesis, and is located almost exclusively in the periplasm.
