چکیده

Changes in intracellular Ca2 concentrations (ICa2 ]j) in platelets stimulated with aggregating agents were measured with the fluorescent indicator dye quin 2. Ca2 influx, but not intracellular mobilization, in response to adenosine diphosphate (ADP). platelet aggregating factor (PAFacether). and sodium arachidonate was significantly inhibited by monoclonal antibodies against the glycoprotein (GP) llb-Illa complex; inhibition of thrombin-stimulated influx was inhibited to a lesser extent and reached statistical significance only at thrombin concentrations of 0.1 U/mI

Fibrinogen Competes With von Willebrand Factor for Binding to the Glycoprotein lib/Illa Complex When Platelets Are Stimulated With Thrombin

Two monoclonal antibodies-one that blocks ristocetininduced platelet binding of von Willebrand factor to glycoprotein lb and one that blocks adenosine diphosphateinduced binding of fibrinogen to the glycoprotein llb/Illa complex-were used to assess the binding site(s) for von Willebrand factor when platelets are stimulated with thrombin or adenosine diphosphate (ADP). Neither agonist induced bi...

Isolation and Structural Characterization of the Polypeptide Subunits of Membrane Glycoprotein lib-Illa From Human Platelets

We have previously demonstrated the isolation of platelet membrane glycoprotein lIb-Illa by affinity chromatography with a specific monoclonal antibody.1 We have now separated the polypeptide subunits lIb and lIla of the isolated glycoprotein by preparative sodium dodecyl sulfate polyacrylamide gel electrophoresis and have compared their structural features. Both llb and lIla contain -.1 5% car...

Platelet Glycoproteins LIb and Lila Associated With Blood Monocytes Are Derived From Platelets

Platelet glycoprotein lIb/Illa (GP lIb/Illa). the receptor complex for fibrinogen, has been regarded as a megakaryocyte/platelet lineage-restricted antigen. Recently. however. it has been reported that GP lIb/Illa is expressed in blood monocytes. Studies were performed to establish the origin and immunological characteristics of monocyteassociated glycoproteins lIb and lIla (GPs lIb and lIla). ...

Exposure of Platelet Fibrinogen-Binding Sites by Collagen, Arachidonic Acid, and ADP: Inhibition by a Monoclonal Antibody to the Glycoprotein lib-Illa Complex

Following stimulation with adenosine diphosphate (ADP). collagen. or arachidonic acid. unstirred human platelet suspensions bind ‘25l-fibrinogen in a reaction that reaches completion within 30 mm. Scatchard analysis of these binding data reveals two sets of binding sites with all 3 agents: a high affinity site (Kd 0.029-0.045 MM) binding 1 000-i 600 fibrinogen molecules per platelet. and a lowe...

von Willebrand Factor Is Present on the Surface of Platelets Stimulated in Plasma

von Willebrand factor (vWf) can bind to glycoprotein (GP) lIb/Illa on activated platelets. The significance of this interaction is unclear, however. because it has not been possible to detect vWf binding to GPIIb/llla on platelets stimulated in plasma. We have developed an indirect. flow cytometry assay that uses fluorescein-labeled antibodies to detect vWf and fibrinogen on platelets. Using th...
