Significant Effect of Hydromechanical vs Mechanical Stimulation on Collagen Production of a Chondrocyte-seeded Hydrogel

چکیده
Introduction: Mechanical stimulation has an effective role in helping to produce more functional engineered articular cartilage tissue for focal defect repair [1]. It is hypothesized that adding direct pressurized fluid flow to the combination of compression and sliding shear acting on chondrocyteseeded hydrogel constructs can significantly increase collagen production. If so, this “hydromechanical” stimulation approach could eventually be applied to cartilaginous tissue that is formed in vitro for cartilage repair.

Evaluating the effect of pH on mechanical strength and cell compatibility of nanostructured collagen hydrogel by the plastic compression method
Objective(s): One of the main constraints of collagen hydrogel scaffolds for using in tissue engineering is mechanical weakness. Plastic compression (PC) is a physical method to overcome the mechanical limitation of collagen hydrogel. Materials and Methods: In this study, the effects of pH on mechanical and biological properties of PC hydrogels were investigated. Collagen hydrogels were fabrica...

Introduction: Natural biomaterials and growth factors are key factors in tissue engineering. The objective of the present study was to evaluate transforming growth factor beta 1 (TGF-β1) and curcumin on proliferation and differentiation of nasal-derived chondrocyte seeded on the fibrin glue scaffold. Methods: Chondrocytes were isolated from nasal samples. Nasal-derived chon...
متن کاملInterstitial Flow Produces a Superficial Zone-Like Layer in Tissue Engineered Cartilage
INTRODUCTION: Surgical repair of articular cartilage typically yields fibrocartilage that lacks the stratified ECM architecture of native cartilage and does not integrate with the surrounding hyaline cartilage. Because of these limitations, there has been considerable interest in cartilage tissue engineering and that led to the development of various bioreactor systems to grow functional engine...
متن کاملA hydrogel-mineral composite scaffold for osteochondral interface tissue engineering.
Osteoarthritis is the leading cause of physical disability among Americans, and tissue engineered cartilage grafts have emerged as a promising treatment option for this debilitating condition. Currently, the formation of a stable interface between the cartilage graft and subchondral bone remains a significant challenge. This study evaluates the potential of a hybrid scaffold of hydroxyapatite (...
متن کاملStudy of Human Chondrocyte Redifferntiation Capacity in Three-Dimensional Hydrogel Culture
Objective(s) Articular cartilage tissue defects cannot be repaired by the proliferation of resident chondrocytes. Autologous chondrocyte transplantation (ACT) is a relatively new therapeutic approach to cover full thickness articular cartilage defects by in vitro grown chondrocytes from the joint of a patient. Therefore, we investigated the redifferentiation capability of human chondrocytes ma...
