چکیده

ATP-binding cassette (ABC) transporters consist of a family of proteins that translocate substrates against a concentration gradient from the intracellular toward the extracellular milieu. Among the members of the ABC transporter superfamily, P-glycoprotein has been studied most extensively and has been found to be expressed in tissues such as liver, kidneys, and intestines. This suggests a physiological function for this transporter in the protection and detoxification of the organism. P-glycoprotein and other ABC transporters were also found to be expressed in the myocardium. Current literature suggests that ABC transporters have a role in controlling distribution of xenobiotics to the heart, thus protecting this organ. Heart Metab. 2007;35:1–6.

The role of ATP-binding cassette transporter A2 in childhood acute lymphoblastic leukemia multidrug resistance

Acute lymphoblastic leukemia (ALL) is one of the most prevalent hematologic malignancies in children. Although the cure rate of ALL has improved over the past decades, the most important reason for ALL treatment failure is multidrug resistance (MDR) phenomenon. The current study aims to explain the mechanisms involved in multidrug resistance of childhood ALL, and introduces ATP-binding cassette...

The ATP-binding cassette transporters and their implication in drug disposition: a special look at the heart.

The passage of drugs across cell membranes dictates their absorption, distribution, metabolism, and excretion. This process is determined by several factors including the molecular weight of the compounds, their shape, degree of ionization, and binding to proteins. Accumulation of xenobiotics into tissues does not depend only on their ability to enter cells, but also on their ability to leave t...

مکانیسم مقاومت دارویی در سرطان

Some varieties of human cancers become resistant, or, are intrinsically resistant to treatment with conventional drug therapies. This phenomenon is due largely to over-expression of the ATP binding cassette, (ABC), super-family of membrane transporters. In this regard, 170 kDa plasma membrane ATP-dependent pump, known as P-glycoprotein are the most important. Other members of multi-drug resista...

Effects of Salinispora derived metabolites against multidrug resistance, an in-silico study

Background: Multidrug resistance (MDR) is known to defeat most chemotherapies as one of the main anticancer strategies. The role of overexpression/overactivation of ABC transporters, especially P-glycoprotein (P-gp), in the development of chemotherapy has long been demonstrated. Salinispora is a marine actinomycete genus known for the production of novel bioactive metabolites. Methods: In this...

O6-Methylguanine-DNA Methyltransferase and ATP-Binding Cassette Membrane Transporter G2 Promotor Methylation: Can Predict the Response to Chemotherapy in Advanced Breast Cancer?

Background: ATP-binding cassette membrane transporter G2 (ABCG2) gene is one of transporter family and well characterized for their association with chemoresistance. Promoter methylation is a mechanism for regulation of gene expression. O6-Methyl guanine DNA methyl transferase (MGMT) gene plays a fundamental role in DNA repair. MGMT has the ability to remove alkyl adducts from DNA at the O6 pos...
