Sarcoplasmic Reticulum Luminal [Ca

چکیده
The role that sarcoplasmic reticulum (SR) luminal Ca 2+ concentration plays in regulating ryanodine receptors (RyRs) is considered to be very important in process of cardiac excitation-contraction coupling. Here we developed a multi-scale mathematical model by coupling a two-dimensional spatio-temporal Ca 2+ reaction-diffusion model with an action potential model of the ventricular myocyte. The simulation results showed that 1) the frequency of spontaneous sparks increased when SR Ca 2+ content was elevated; 2) when SR Ca 2+ was overloaded, spontaneous Ca 2+ waves might occur without any external stimulus, and 3) once formed, the propagation of Ca 2+ waves was accelerated as the SR Ca 2+ content increased. Moreover, those spontaneously occurred Ca 2+ release events elevated cytoplasmic [Ca 2+ ]i and then activated inward currents which might cause a delayed afterdepolarization (DAD).

Ryanodine receptor luminal Ca2+ regulation: swapping calsequestrin and channel isoforms.
Sarcoplasmic reticulum (SR) Ca(2+) release in striated muscle is mediated by a multiprotein complex that includes the ryanodine receptor (RyR) Ca(2+) channel and the intra-SR Ca(2+) buffering protein calsequestrin (CSQ). Besides its buffering role, CSQ is thought to regulate RyR channel function. Here, CSQ-dependent luminal Ca(2+) regulation of skeletal (RyR1) and cardiac (RyR2) channels is exp...

Changes in cytoplasmic Ca-levels regulate the contractile status of skeletal muscle fibres, whereby the finely tuned interplay between voltage sensors, Ca-release channels, Cabinding proteins and Ca-pumps mediates Ca-cycling through the sarcoplasmic reticulum. Although the physical coupling between the α1S-dihydropyridine receptor of the transverse tubules and the ryanodine receptor Ca-release ...
متن کاملCardiac ryanodine receptor activation by a high Ca2+ store load is reversed in a reducing cytoplasmic redox environment
Here, we report the impact of redox potential on isolated cardiac ryanodine receptor (RyR2) channel activity and its response to physiological changes in luminal [Ca(2+)]. Basal leak from the sarcoplasmic reticulum is required for normal Ca(2+) handling, but excess diastolic Ca(2+) leak attributed to oxidative stress is thought to lower the threshold of RyR2 for spontaneous sarcoplasmic reticul...
متن کاملControl of cardiac ryanodine receptor by sarcoplasmic reticulum luminal Ca2+
Introduction Cardiac myocyte contraction is driven by the coordinated release of Ca from the sarcoplasmic reticulum (SR). This release of Ca occurs through the cardiac RyR2. Physiologically, Ca release occurs in response to an influx of Ca through L-type Ca channels, via a mechanism termed CICR. This transient increase in cytosolic Ca activates the RyR2 channel as a result of Ca binding to the ...
متن کاملEvidence for Ca Activation and Inactivation Sites on the Luminal Side of the Cardiac Ryanodine Receptor Complex
We have used tryptic digestion to determine whether Ca can regulate cardiac ryanodine receptor (RyR) channel gating from within the lumen of the sarcoplasmic reticulum (SR) or whether Ca must first flow through the channel and act via cytosolically located binding sites. Cardiac RyRs were incorporated into bilayers, and trypsin was applied to the luminal side of the bilayer. We found that befor...
