چکیده
Background: Caldecrin is a serum calcium-decreasing factor and inhibits osteoclast differentiation. Results: Caldecrin inhibits RANKL-stimulated Ca 2+ entry, sealing actin ring formation and bone resorption of mature osteoclasts in a c-Src-dependent manner. Conclusion: Caldecrin is a negative regulator of RANKL-mediated osteoclast function. Significance: Caldecrin contributes to the understanding of the unique regulatory system for c-Src in osteoclasts.

متن کاملProtein kinase N3 promotes bone resorption by osteoclasts in response to Wnt5a-Ror2 signaling.
Cytoskeletal reorganization in osteoclasts to form actin rings is necessary for these cells to attach to bone and resorb bone matrices. We delineated the pathway through which Wnt5a signaling through receptor tyrosine kinase-like orphan receptor 2 (Ror2) promoted the bone-resorbing activity of osteoclasts. Wnt5a binding to Ror2 stimulated Rho, a small GTPase involved in cytoskeletal reorganizat...
متن کاملAnti-inflammatory effect of Yu-Ping-Feng-San via TGF-β1 signaling suppression in rat model of COPD
Objective(s): Yu-Ping-Feng-San (YPFS) is a classical traditional Chinese medicine that is widely used for treatment of the diseases in respiratory systems, including chronic obstructive pulmonary disease (COPD) recognized as chronic inflammatory disease. However, the molecular mechanism remains unclear. Here we detected the factors involved in transforming growth factor beta 1 (TGF-β1)/Smad2 si...
متن کاملInhibition of Osteoclast Activation by Phloretin through Disturbing αvβ3 Integrin-c-Src Pathway
This study was to explore the sequential signaling of disorganization of the actin cytoskeletal architecture by phloretin. RAW 264.7 macrophages were incubated with 1-20 μM phloretin for 5 days in the presence of RANKL. C57BL/6 mice were ovariectomized (OVX) and orally treated with 10 mg/kg phloretin once a day for 8 weeks. Phloretin allayed RANKL stimulated formation of actin podosomes with th...
متن کاملInvolvement of the Src-cortactin pathway in podosome formation and turnover during polarization of cultured osteoclasts.
Osteoclasts are large, multinucleated cells that adhere to bone via podosomes, and degrade it. During osteoclast polarization, podosomes undergo reorganization from a scattered distribution, through the formation of clusters and ring super-structures, to the assembly of a sealing zone at the cell periphery. In the present study, we demonstrate that the levels of podosome-associated actin, and i...
