The synthesis of heme and globin in the maturing human erythroid cell.

چکیده
This report describes observations on the rate of heme and globin synthesis within the maturation period of human marrow erythroid cells. Such studies have been designed to examine the hypothesis that heme and globin are synthesized with temporal independence during erythroid cell maturation and to establish the normal relationship of heme and globin synthesis. With such data it is hoped that further work may lead to a definition of some of the effects of certain hormones and drugs upon heme and globin synthesis and to closer examination of certain refractory anemias with particular emphasis on abnormalities of globin synthesis. The rates of synthesis of heme and globin within the erythroid cell maturation period are defined in these studies by the daily incremental incorporation of glycine-2-C14 into peripheral blood hemin and globin and of Fe59 into red cells following intravenous administration of these precursors to four healthy male volunteers.

Erythroid Ce11 Differentiation
The study of erythroid cell difierentiation is pertinent to the study of human leukemia from two points of view. First, since a common stem cell gives rise to both erythroid and myeloid cells, it is not unexpected that disorders of myeloid cell proliferation and differentiation should be occasionally associated with abnormalities of erythroid cell differentiation. In fact in many cases of human...
متن کاملIn vitro Induction of Fetal Hemoglobin in Erythroid Cells Derived from CD133 Cells by Transforming Growth Factor-b and Stem Cell Factor
Increased fetal hemoglobin (HbF) in b-globin gene disorders ameliorates the clinical symptoms of the underlying disease. 5-azacytidine, butyrate and hydroxyurea, have been shown to activate g-globin gene expression. It has also been found that hematopoietic growth factors can influence expression of g-globin in erythroid cultures and in animal models. This study was designed to evaluate the in ...

Background and aim: Induction of fetal hemoglobin (Hb-F) can improve the patients’ symptoms of haemoglobinopathies. Several factors can induce gamma globin gene expression and increased Hb-F levels in patients. In this study, the expression of genes is involved in regulation of gamma globin synthesis such as PIPKII-alpha BCL11a, and miR-30a during CD34+ hematopoietic stem cell differentiation i...
متن کاملDeficient Heme and Globin Synthesis in Embryonic Stem Cells Lacking the Erythroid-Specific d-Aminolevulinate Synthase Gene
The erythroid-specific isoform of d-aminolevulinate synthase (ALAS-E) catalyzes the first step of heme biosynthesis in erythroid cells, and ALAS-E gene mutations are known to be responsible for x-linked sideroblastic anemia. To study the role of ALAS-E in erythroid development, we prepared mouse embryonic stem (ES) cells carrying a disrupted ALAS-E gene and examined the effect of the lack of AL...
متن کاملIntracellular heme coordinately modulates globin chain synthesis, transferrin receptor number, and ferritin content in differentiating Friend erythroleukemia cells.
The effect of succinylacetone (SA), a highly specific inhibitor of ALA-dehydratase and heme synthesis, on hemoglobin (Hb) production, transferrin receptor (TfR), and ferritin expression was analyzed in differentiating Friend leukemia cells (FLC). This compound exerted a pronounced inhibitory effect not only on heme and Hb synthesis, but also on all the remaining above-mentioned parameters. In p...
